			<content:encoded><![CDATA[<p>Ipilimumab (anti-CTLA-4) foi aprovado pela agencia americana <a href="http://www.fda.gov">FDA</a> para o tratamento do melanoma metasta&#8217;tico, nao ressecavel.</p>
<p><a href="http://www.bms.com/pages/default.aspx">Ipilimumab</a> e&#8217; um tratamento passivo immuno-terapeutico que usa anticorpos blocantes da interaction de CTLA-4 expresso na superficie dos linfo&#8217;citos T, com mole&#8217;culas costimuladoras como CD80 e CD86.</p>
<p><a href="http://www.ncbi.nlm.nih.gov/pmc/articles/PMC2714548/?report=abstract&amp;tool=pmcentrez">CTLA-4</a> e&#8217; expresso quando as ce&#8217;lulas T sao activadas competindo com CD28 para a ligacao com CD80 e CD86. Ao faze-lo leva ao blocamento do sinal de costimulacao e frena a activacao celular do linfocito, reduzindo a sua actividade funcional.</p>
<p>Ipilimumab como antagonista de CTLA-4 impede este sistema de autocontrolo negativo dos linfocitos permitindo que os linfo&#8217;citos continuem a manter o seu estado funcional, conduzindo por isso a uma mais eficaz accao sobre as ce&#8217;lulas tumorais.Resultados cli&#8217;nicos de<a href="http://www.ncbi.nlm.nih.gov/pubmed/19001147"> fase II e III</a> foram muito encoragedores levando &#8216;a aprovacao pelo FDA.</p>
<p>No entanto, a accao de ipilimumab nao e&#8217; inocua, pois tambe&#8217;m leva a que os processos de controlo de accao de linfo&#8217;citos se torne descontrolado sobretudo em situacoes inflamatorias e induzindo respostas autoimmunes nao neglijaveis.</p>
<p>Apo&#8217;s a aprovacao de Provenge pela FDA para o tratamento do cancro da prostata, Ipilimumab e&#8217; o segundo producto imunoterapeutico aprovado para o tratamento do cancro.</p>

			<content:encoded><![CDATA[<p>E se o cancro for o resultado de uma catástrofe ao nível dos cromossomas em vez de um processo linear, progressivo de mutações aleatórias no genoma?</p>
<p>Um artigo recentemente publicado no jornal <a title="Massive Genomic Rearrangement Acquired in a Single Catastrophic Event during Cancer Development (Stephens et al.)" href="http://www.cell.com/abstract/S0092-8674(10)01377-2">Cell</a>, demonstra que a célula neoplásica resulta provavelmente de uma drástica fragmentação dos cromossomas de uma célula normal com formação aleatória de vários segmentos de um cromossoma, que são depois reunidos de forma anormal constituindo um novo cromossoma, pelos mecanismo de reparação do ADN das células afectadas.</p>

<p>Por consequência, esta reorganização genómica resultara na mutação de oncogenes (que promovem o cancro), deleção de genes onco-supressores (que reprimem ou impedem o cancro) permitindo o desenvolvimento neoplásico.</p>

			<content:encoded><![CDATA[<p>Num artigo publicado na revista <a href="http://www.sciencemag.org/content/early/2011/01/12/science.1200801">science</a>, é descrito que a  detecção da translação de sequências microsatélite nas células cancerígenas podem ser usado como marcador molécular.</p>
<p>Estas sequencias microsatélite são já usadas para o diagnóstico do cancro a partir da sequenciação de regiões do genoma, mas deconhecia-se que as mesmas sequências poderiam ter ser transcritas, facto demonstrado no artigo serem particulares do cancro e ausentes nas células adultas normais.</p>
<p>Este artigo vem abrir novas prespectivas de diagnóstico precoce ou monitorização do mesmo em situações clinicas ou sem sintomas clinicos.</p>

<p>Os resultados foram demonstrados recentemente na conferencia <a href="http://chicago2010.asco.org">ASCO </a> (American Society of Clinical Oncology) em Chicago.</p>
<p>Os resultados demonstram que o anticorpo blocante (ipilimumab) num estudo clínico de fase III,  que o tratamento isolado ou associado a uma vacinacao com gp100 (um antigénio de defirenciação melanocítica) demonstra um benefício de 4 meses na sobrevida media dos doentes, em comparação com doentes somente tratados com gp100.</p>

<p>O filme mostra linfócitos (verde) a percorrer capilares no cerebro de rato (a vermelho) através de uma técnica chamada <a href="http://www.microscopyu.com/articles/fluorescence/multiphoton/multiphotonintro.html">Two-Photon microscopy</a>. Notar que certos linfocitos atravessam o endotélio e visitam o tecido adjacente em condições experimentais de esclerose multipla.</p>

<p>Evidências de trogocitose começam a referidas em várias <a title="Link a Pubmed (trogocytosis &amp; immune)" href="http://www.ncbi.nlm.nih.gov/sites/entrez?db=pubmed&amp;cmd=DetailsSearch&amp;term=trogocytosis%5Btiab%5D+AND+immune%5Btiab%5D&amp;log$=activity">publicações científicas</a>.</p>
<p>Ras é um comum oncogene, que quando mutado induz tumorigenese.  Numa recente publicação do jornal <a href="http://www.plosone.org">PLoS ONE</a> (de livre acesso), é <a href="http://www.plosone.org/article/info%3Adoi%2F10.1371%2Fjournal.pone.0001204#abstract0">descrito</a> que H-RAS é adquirido via trogocitose pelas células NK e células T quando contactam com células tumorais. A acquisição de H-RAS pelos linfócitos, induz nestes a activação de  ERK via a sua fosforilação, a produção de Interferão-gamma e TNF (&#8220;<em>tumor necrosis factor</em>&#8220;)-  alpha (duas citocinas fundamentais nos processos inflamatórios e actividade lítica anti-tumoral). O resultado final é um potenciamento da activade anti-tumoral dos linfócitos T e NK.</p>
<p>Estas evidências vem aumentar o interesse pelo fenómeno de trogocitose na immunologia tumoral e permitir o desenvolvimento de estratégias de manipulação e identificação de células com actividade anti-tumoral.</p>

			<content:encoded><![CDATA[<p>EU considero que esta procissão de dias disto e daquilo é uma grande estupidez imposta à força por injecções publicitárias com fins comerciais, e admito que Natal não seria Natal se não houvesse por detrás o interesse económico, e talvez por isso é que o presépio é substituido pelo pinheiro depenado assente sobre uma pilha de cartões multicolores com prendas&#8230; como que a dizer que o Natal é sinónimo de prendas e não da celebração do nascimento de Cristo e de tudo que dai advém.</p>
<p>Mas voltando à temática que me leva a escrever esta pequena crónica é que  deparei-me com a<a href="http://www.eurekalert.org/pub_releases/2009-02/uol-clc021309.php"> interessante informação</a> que a maioria das rosas vendidas no período da doideira consumista disfarçada de amor partilhado e saido da gaveta na data apropriada, provêem sobretudo de um pais Africano, o Quénia (potência mundial produtora de rosas vermelhas), e vendidas em leilão no mercado de Amesterdão na Holanda. Ora devido a este consumo de rosas , o pobre lago Naivasha tem desaparecido a olhos vistos estando a sua persistência em risco devido ao consumo desenfreado de água para a produção dos belos rebentos longilíneos rematados de um botão tímido e efémero.</p>

<p>Acho que é bastante estúpido por razões consumistas e publicitárias pôr em risco um ecossistema e a persistência de uma recurso fundamental à subsistência de uma região.</p>
<p>Portanto abstenham-se destas euforias da moda e se sim o desejem plantem lá em casa num vaso a vossa rosa. De certo que quando doada à pessoa bem amada ela terá um valor bem mais interessante e sentimental que algo crescido em série e extensivamente</p>
