What is HPV?

Bowel cancer test research

Australian researchers have developed gene expression biomarkers which can accurately discriminate pre-cancerous and cancerous colorectal growths from non-cancerous controls.

Being presented today at the Digestive Disease Week conference in Chicago, the preliminary findings are the result of a collaborative study involving CSIRO, Flinders University and Australian healthcare company, Clinical Genomics Pty Ltd designed to develop an improved screening/diagnostic test for detecting bowel cancer and significant pre-cancer lesions.

"If we can now show that the levels of these biomarkers in blood or stool also correlate strongly with disease state in a large group of patients with cancer or pre-cancer lesions (ie adenomatous polyps) we may have the basis for a very important new diagnostic weapon in the fight against bowel cancer," says CSIRO's Preventative Health National Research Flagship Theme Leader in Colorectal Cancer and Gut Health, Dr Trevor Lockett.

According to Professor Graeme Young from Flinders University's Centre for Cancer Prevention and Control, regular screening for bowel cancer in people aged 50 years and over is a powerful tool for reducing the impact of the disease in Australia.

"If we can develop a screening test that can point to the presence of clinically important pre-cancerous adenomas which are then removed during follow-up colonoscopy, we will actually be able to prevent the occurrence of bowel cancer in some cases," Professor Young says.

Genomic Health Study Shows Breast Cancers in Men Display Very Similar Gene Signatures to Those in Women

ORLANDO, Fla., May 29, 2009 /PRNewswire-FirstCall via COMTEX/ ----Genomic Health, Inc. (Nasdaq: GHDX) today announced results from a study which summarized the gene signatures identified by the Oncotype DX(R) breast cancer test in a large number of male patients for whom the test was used to guide treatment with chemotherapy. The results, which will be presented in a poster presentation on Monday, June 1 (1:00 - 5:00 p.m. ET) at the American Society of Clinical Oncology (ASCO) annual meeting in Orlando, demonstrated that breast cancer in men displays similar gene signatures to female breast cancer.

"Breast cancer is not just a woman's disease, yet treatment for men is typically extrapolated from our experience in the female population since it is rare and there is little known about the biology of male breast cancer," said George Sledge, M.D., Ballve-Lantero Professor of Oncology at Indiana University in Indianapolis, and lead investigator of the study. "This is the largest genomic study to compare gene expression in female and male patients, and results support the clinical utility of Oncotype DX in providing quantitative information to help guide treatment decisions for men with breast cancer."

The study, "Molecular characterization of male breast cancer by standardized quantitative RT-PCR analysis: First large genomic study of 347 male breast cancers compared to 82,434 female breast cancers" (Abstract #549), analyzed quantitative gene expression by gender status in estrogen receptor (ER) positive tumor specimens. As with female breast cancer, a wide inter-patients variation was observed in gene expression in male breast cancer. The proportion of tumors with low risk of recurrence based on the Recurrence Score(R) was 53.6 percent in males versus 53.4 percent in females, intermediate risk of recurrence was 35.2 percent in males versus 36.3 percent in females, and high risk of recurrence was 11.2 percent in males versus 10.3 percent in females.

Despite the similarities between the two patient groups, there were also some differences. For example, male breast cancer patients had higher mean expression of the hormone receptor genes, likely due to the different hormonal context of men and women. In addition, male breast cancer patients were older, on average, and were less likely than women to have the lobular form of breast cancer.

"I was quite surprised when my doctor told me I had breast cancer and was not looking forward to the multiple rounds of chemotherapy that he originally recommended," said Mike Nelsen, a breast cancer patient from Baltimore. "After my Oncotype DX test results came back and I discussed them with my oncologist, I felt confident that I could avoid chemotherapy and its side effects without increasing the risk of my cancer returning."

Multiple Studies Highlight Application of Personalized Medicine in Oncology Setting

Additionally, Genomic Health, in partnership with investigators from several academic institutions, will present a separate study at ASCO evaluating the genotypic features of triple-negative breast cancer compared with hormone receptor-positive disease, as well as the genotypic features associated with recurrence. The study, "Genotypic characterization of phenotypically defined triple-negative breast cancer" (Abstract #500), demonstrated that there were significant differences in gene expression between the triple negative and hormone receptor positive groups, including genes for which targeted agents are currently being evaluated in the clinical environment. Full results will be discussed in an oral presentation on Sunday, May 31 (4:30 p.m. ET).

These presentations are in addition to Genomic Health's previously announced results regarding its Oncotype DX colon cancer test, from the landmark QUASAR validation study, demonstrating that the test can independently predict individual recurrence risk in stage II colon cancer patients following surgery. The study, "A quantitative multi-gene RT-PCR assay for prediction of recurrence in stage II colon cancer: Selection of the genes in 4 large studies and results of the independent, prospectively-designed QUASAR validation study" (Abstract #4000), which was highlighted on May 14 during ASCO's advance presscast, will be presented Saturday, May 30 (3:00 p.m. ET). FDA Approves First Canine Cancer Drug

The Food and Drug Administration approved today the nation’s first drug developed specifically for the treatment of canine cancer.

Manufactured by Pfizer Animal Health Inc., based in New York City, the drug marketed as Palladia, or chemically known as toceranib phosphate, will hit the markets in early 2010.“This cancer drug approval for dogs is an important step forward for veterinary medicine,” Bernadette Dunham, D.V.M., Ph.D., director of FDA’s Center for Veterinary Medicine, said in a released statement.

“Prior to this approval, veterinarians had to rely on human oncology drugs, without knowledge of how safe or effective they would be for dogs. Today’s approval offers dog owners, in consultation with their veterinarian, an option for treatment of their dog’s cancer.”

Current cancer drugs used in veterinary medicine are in fact not approved for use in animals, as they were originally designed for human use. But vets are allowed to administer human cancer medicine and other treatments under Federal law for controlled substances, per Animal Medicinal Drug Use Clarification Act of 1994.

Before Palladia’s mainstream release, Pfizer plans to introduce Palladia to boarded specialists to expand the body of clinical experience with the United States’ first canine cancer therapy.

“At the completion of a Palladia clinical study, approximately 60 percent of dogs had their tumors disappear, shrink or stop growing,” said Cheryl London, who has helped Pfizer Animal Health’s Veterinary Medicine Research & Development to develop Palladia since 2000.

Palladia is a tyrosine kinase inhibitor and works in two ways: by killing tumor cells and by cutting off the blood supply to the tumor. In a clinical trial, Palladia showed a statistically significant difference in tumor shrinkage when compared with the placebo.

The first-of-its-kind drug could vastly change the lives of dogs as cancer is the leading cause of death in canines, according to the Morris Animal Foundation. Additionally, Pfizer estimates 1.2 million new canine cases are reported in the U.S. annually.

Palladia tablet is indicated to treat Patnaik grade II or III recurrent cutaneous mast cell tumors with or without regional lymph node involvement. This type of cancer, the FDA reports, is responsible for one-out-of-five case of canine skin tumors.

While canine mast cell tumors often appear small and insignificant, they can be a very serious form of cancer in dogs. Some mast cell tumors are easily removed without the development of any further problems, while others can lead to life threatening disease.

Palladia is approved to treat the tumors with or without regional lymph node involvement. Common side effects include diarrhea, decrease or loss of appetite, lameness, weight loss, and blood in the stool.

Many postmenopausal women are looking for alternatives to hormone therapy, especially in light of the recent Women’s Health Initiative research findings concerning the risks of combined estrogen and progestin therapy. Of particular interest are phytoestrogens, which have been gaining popularity due to their “natural” status, alleged health claims, and availability in a wide range of foods and supplements.

What are Phytoestrogens?

Phytoestrogens are naturally occurring plant compounds that have some similarities to estradiol, the most potent naturally occurring estrogen. However, phytoestrogens tend to have weaker effects than most estrogens, are not stored in the body, and can be easily broken down and eliminated.

Observational studies have found a lower prevalence of breast cancer, heart disease and hip fracture rates among people living in places like Southeast Asia, where diets are typically high in phytoestrogens. In North America, knowledge of these reported health effects has stimulated great interest in the health benefits of phytoestrogens. According to the Food and Drug Administration, the sale of soy foods, a major source of phytoestrogens, has increased dramatically in the past decade.

Dietary Sources of Phytoestrogens

Phytoestrogens consist of more than 20 compounds and can be found in more than 300 plants, such as herbs, grains and fruits. The three main classes of dietary phytoestrogens are isoflavones, lignans and coumestans:

1. Isoflavones (genistein, daidzein, glycitein and equol) are primarily found in soy beans and soy products, chickpeas and other legumes.

2. Lignans (enterolactone and enterodiol) are found in seeds (primarily flaxseed), cereal bran, legumes, and alcohol (beer and bourbon).

3. Coumestans (coumestrol) can be found in alfalfa and clover. Most food sources containing these compounds typically include more than one class of phytoestrogens.

The Skeletal Effects of Phytoestrogens

Much of the evidence concerning the potential role of phytoestrogens in bone health is based on animal studies. In fact, soybean protein, soy isoflavones, genistein, daidzein and coumestrol have all been shown to have a protective effect on bone in animals who had their ovaries surgically removed.

In humans, however, the evidence is conflicting. Compared to Caucasian populations, documented hip fracture rates are lower in countries such as Hong Kong, China and Japan where dietary phytoestrogen intakes are high. Yet reports suggest that Japanese women have a greater risk of sustaining a vertebral fracture than Caucasian women.

Several studies have explored the effects of soy isoflavones on bone health, but results have been mixed, ranging from a modest impact to no effect. Most of these studies have serious limitations, including their short duration and small sample size, making it difficult to fully evaluate the impact of these compounds on bone health.

Ipriflavone Supplements

Ipriflavone, a synthetic isoflavone, has shown some promise in its ability to conserve bone in postmenopausal women. Ipriflavone has also been shown to have a protective effect on bone density in pre-menopausal women taking gonadotropin-releasing hormone (GnRH), a treatment for endometriosis that triggers bone loss.

However, a definitive three-year study of more than 400 postmenopausal women concluded that ipriflavone did not prevent bone loss. Additionally, the compound was linked to lymphocytopenia (a reduction in lymphocytes) in a significant number of study participants. Lymphocytes are a type of white blood cell that helps the body fight infection.

Risks and Benefits Are Unclear

Some studies suggest that, unlike estrogen, phytoestrogens do not appear to target breast or uterine tissue. This suggests that they may act more like SERMS (selective estrogen receptor modulators such as raloxifene and tamoxifen) than actual estrogens. However, in other studies high isoflavone levels have been linked to an increased risk of breast cancer.

Clearly, additional research is needed to further evaluate the effects of phytoestrogens before judgments regarding their safety and usefulness can be made.

Key Points

Based on information available at this time, it is reasonable to make the following conclusions concerning phytoestrogens and bone health in postmenopausal women:

1. Moderate amounts of foods containing phytoestrogens can be safely included in the diet but do not expect it to help build bone. Keep to the basic rule - eat the least processed forms.

2. Due to a lack of evidence and concerns about safety, supplementation with synthetic isoflavones (ipriflavone) is in question.

3. Postmenopausal women are encouraged to view evidence concerning phytoestrogens and bone health as conflicting and incomplete. For women who are estrogen dominant increasing their phytoestrogen intake may not improve their bone position.

TGen partners with John C. Lincoln Health Network to prevent breast cancer

PHOENIX, Ariz. – May 4, 2009 – Today's opening of a new breast health center next to John C. Lincoln Deer Valley Hospital will provide significant research opportunities for the Translational Genomics Research Institute (TGen).

The 9,000-square-foot Breast Health and Research Center will include a tumor biorepository for TGen that will aid the non-profit research institute in discovering new ways to diagnose and treat breast cancer, which affects 1 in every 8 American women.

"What this partnership means is that breast cancer research at TGen will be significantly accelerated, specifically in development of more accurate diagnostic tests and smarter treatment options for newly-diagnosed breast cancer patients,'' said Dr. Heather Cunliffe, head of TGen's Breast & Ovarian Cancer Research Unit.

The new center is located in John C. Lincoln's Medical Offices 3, 19646 N. 27th Ave., just southwest of Interstate 17 and Loop 101, in northwest Phoenix.

Tips For Safe Meat Grilling (Cut Cancer Risks)

The American Institute of Cancer Research is urging everyone to rethink the pastime of barbecuing meat.

After analyzing the results of 7,000 studies, the Institute concluded that grilling any meat — whether red, white or fish — produces potent carcinogens.

The high heat of

grilling reacts with proteins in red meat, poultry and fish, creating heterocyclic amines, which are linked to cancer. Another form of cancer-causing agents, polycyclic aromatic hydrocarbons, are created when juices from meats drip and hit the heat source. They then rise in smoke and can stick to

the meat.

The Institute took particular aim at processed meats like hot dogs, sausages, bacon, ham, pastrami, salami and any meat that has been salted, smoked or cured. The chemicals used to preserve the meat increase the production of cancer-causing compounds, regardless of how

the meat is cooked.

Any time you cook meat at high

temperatures, whether you’re
grilling, frying, broiling, etc., some pretty nasty chemicals are created:

Heterocyclic Amines (HCAs): These form when food is cooked at high

temperatures, and they’re linked to cancer. In terms of HCA, the worst part of
the meat is the blackened section, which is why you should always avoid charring your meat, and never eat blackened sections.

Polycyclic

Aromatic Hydrocarbons (PAHs): When fat drips onto the
heat source, causing excess smoke, and the smoke surrounds your food, it can transfer cancer-causing PAHs to
the meat.

Advanced Glycation End Products (AGEs): When food is cooked at high temperatures (including when it is pasteurized or sterilized), it increases the formation of AGEs in your food. When you eat the food, it transfers the AGEs into your body. AGEs build up in your body over time leading to oxidative stress, inflammation and an increased risk of heart disease, diabetes and kidney disease.

It’s ironic, isn’t it, that so many people are convinced they must cook their foods until they’re browned and “well-done” to avoid bacteria, when in so doing they are creating and consuming highly toxic substances that will shorten their lifespan?

There are many people who are convinced that being a vegetarian is the ultimate healthy lifestyle eating plan. They support their position with loads of scientific studies. But it is my belief that most of these studies are correct because the common way most people consume animal protein is COOKED, or worse yet grilled, creating all the toxic substances discussed above.

To the best of my knowledge there are no studies comparing raw animal-food-based diets versus cooked vegetarian diets, but even if there were they still could be flawed because they most likely would not factor in the person’s nutritional type.

Remember a carb nutritional type eating large amounts of raw animal food will do just as poorly as a protein type on raw vegan diet. You simply need the right food for your specific nutritional type.

One variable that to consider is that you simply don’t have to be perfect in your diet. I fully believe you just need to eat very well 80-95% of the time. If you do that your body typically has enough reserve to compensate for the damage you cause. However if you are really sick or have a terminal illness you will want to get as close to 100% as practically possible to improve your chances of recovery.

Do You STILL Want to Over-Cook Your Food?

If so, consider the following, quite sobering, statistics:

1. In one study, researchers found that those who ate their beef medium-well or well-done had more than three times the risk of stomach cancer as those who ate their beef rare or medium-rare.

2. Other studies have shown that an increased risk of developing pancreatic, colorectal, and breast cancer is associated with high intakes of well-done, fried, or barbecued meats.

3. One study found that a compound called PhIP, formed when meat is charred at high

temperatures, causes prostate cancer in rats.

4. Scientists have estimated that the average cancer risk because of heterocyclic amine exposure ranges from 1 per 10,000 for the average person to more than 1 per 50 for those ingesting large amounts of well-done muscle meats, especially flame-grilled chicken.

All Meat is NOT Inherently Bad for You

As usual, there are errors in studies like the one above. If you take it at face value, you are left with the incorrect notion that meat is something you should only eat sparingly, if at all. Yet there are numerous factors that influence the quality or health-value of

the meat you eat, even above and beyond how it’s cooked. That includes:

Whether or not it’s organic (conventional meat is loaded with pesticides, hormones, antibiotics and other chemicals)

Whether or not it’s grass-fed (essential for healthy meat)

Whether or not it contains nitrates, preservatives linked to cancer

What it did point out is that processed meats are a big-time no-no, and I have to agree with them there. While you can find natural and organic processed meats, the unprocessed varieties are still preferable.

The other factor that plays a role in how healthy meat is for you? As I said earlier, your nutritional type. Some of you (including myself) thrive on red meat while others need white meats or mostly other protein sources, like eggs. It all depends on your unique biochemistry.

For Those of You Who Won’t Have Summer Without Your Grill

I suspect this may be a great number of you, so I will include some tips to minimize your risk.

1. Limit the amount of grilled foods you eat, and make sure you’re eating plenty of other raw foods in your diet.

2. You can reduce the amount of PAHs when you grill by not cooking fatty meats, and by trimming the fat off before you grill.

3. When

grilling, cook your food with indirect heat, such as on a rack rather than directly on the coals. Cooking on a cedar plank is also helpful.

4. Always avoid charring your meat (and don’t eat the black or brown parts).

5. Cook meat partially before putting it on the grill, or cook smaller pieces of meat, which take less time to cook, and therefore give HCAs less time to form.

6. You can reduce the amount of AGEs in your food by using an acidic marinade that contains lemon juice or vinegar.

7. Marinating meats before
grilling or broiling them can reduce HCAs (according to some experts by 90 percent or more). However, only use natural ingredients for marinades, and keep the coating thin to avoid charring.

8. Flip your burgers often, as this will help cut down on HCAs.

9. Add blueberries or cherries to your burgers, as they can also help prevent the formation of HCAs.

10. Avoid
grilling hot dogs, bratwurst and other processed meats, as these seem to be among the worst offenders.

11. Only grill high-quality, organic and grass-fed meats.

12. Cook
the meat as little as possible. Rare or medium-rare at the absolute most. You can also quickly sear

the meat on both sides, leaving the inside mostly raw. This gives the illusion that you’re eating cooked meat, with many of the benefits of raw.
