<p>In the 30 years since AIDS was first discovered, millions of individuals throughout the world have lost their lives. The scientists and researchers devoted to combating both HIV and AIDS have made incredible progress over the last few decades but there is still no cure or effective vaccine. Antiviral therapy has helped individuals to live normal lives and has greatly reduced the number of AIDS-related deaths throughout the world. HIV is often considered to be a chronic disease rather than one that brings with it an automatic death sentence thanks to the drugs available but an individual will require lifelong therapy. In spite of the many advances of that have been made, there are still troublesome issues that must be resolved.</p>
<p>Pharmacogenomics is a fairly recent science which is focused on individualizing a person&#8217;s therapy in order to decrease toxicities and improve success rates for drug therapy. The pharmacogenomics of HIV therapy helps researchers take into account the genetic diversity of both the virus and the individual. This approach to drug therapy reduces failure rates as well as both the long and short term toxicities of the antiviral drugs used to treat the virus. The current approach to treating HIV and AIDS is intended to prolong the patient&#8217;s life by suppressing viral replication.</p>
<p>Researchers do not agree on exactly when drug therapy should begin and there are profound differences in the way that both physicians and scientists approach treatment. <p>Each person has a unique DNA, determined by a small fraction spread throughout the total genome. Unless you have an identical twin, no one else has the same sequence of bases in all of the sections of DNA.</p>
<p>DNA testing is based particularly on these unique sequences, analyzing these specific sections called markers. DNA samples can be taken from cells found in body fluids, skin, hair follicles or the inside of the cheek. After isolating the DNA from the cells, millions of copies are made using an enzyme that speeds DNA reproduction. Then, other reactions break apart the DNA molecules at specific locations in order to isolate particular markers. These markers are ultimately compared to the unknown sample. The more markers identified, the more chances that the match between two samples is accurate. DNA analysts look at a number of different markers to create a &#8220;DNA fingerprint&#8221;.</p>
<p>All methods of collecting DNA samples, be it blood, cheek swab, skin, is equally accurate, because all cells contain the same DNA. So contrary to popular belief, a blood sample is no more accurate than a cheek swab, for example. However, when hair samples are used, the hair must be pulled, not cut, because hair is not made of living cells. If it &#8216;s pulled it carries with it some cells from the living follicle.</p>
<p>It is impossible to absolutely identify a person as the source of a DNA sample without looking at the whole of that person &#8216;s DNA. With identical twins, it &#8216;s never possible, but with many markers the chances for an accurate identification are favorable. Typically, the comparison is made using between 2 and 13 markers. With 13 or close, it &#8216;s extremely unlikely that an incorrect identification will occur, however it is not absolutely impossible.</p>
<p>Still, if an absolute match is hardly possible, a mismatch is very easy to identify through DNA analysis. For example, in a criminal investigation a suspect can be easily eliminated as the source of a sample based on the absence of a single marker. All DNA from a particular person must match from one end of each molecule to the other.</p>
<p>DNA testing has consisted a huge breakthrough, and is a strong pillar in any criminal investigation. DNA analysis has even been used to reopen old cases that were considered solved, because DNA samples remain useful for a very long time. The results are far more accurate and reliable than any other form of identification, and numerous people have been cleared for crimes they did not commit based on DNA analysis.</p>

<p>PCR or polymerase chain reaction is a process in which you  can quantify your genetic material DNA (Deoxy-ribonucleic acid) or RNA  (Ribonucleic acid). There are various methodologies of PCR that can be  utilised to amplify these genes and make subsequent copies of them.  While reverse transcriptase PCR works on amplifying RNA molecules, Real  time PCR is primarily used to amplify DNA in order to understand them  more by performing various consecutive experiments on them. Now this  QPCR process is being used commercially by various biological  institutes, research organisations and pharmaceutical firms for various  applications.</p>
<p>The way in which real time PCR works is the genetic  sequence that is to be amplified is tagged with a particular short  sequence called a primer which is a short DNA sequence with a  complementary sequence to the DNA of interest. With the help of  particular enzymes, this DNA sequence is duplicated by linking and  creating complementary DNA strands from the primer of the target DNA  with the help of QPCR. This process goes on repeating and then finally  there are multiple copies of this DNA that are created for multiple  applications.</p>

<p>Researchers at the University of Illinois placed 48 obese women into  two groups. One group was given a high protein diet, and the other group  was placed on a diet with an equal number of calories but more  carbohydrates. The women walked many times a week, but one key is that  the &#8220;higher-protein&#8221; group also added in a couple sessions of resistance  exercise.</p>
<p>When the study was over, the group on the protein rich  diet lost 22 pounds on average and the majority of the weight loss was  from fat. The other women on the high carbohydrate diet lost an average  15 pounds, but an average of two pounds of the weight they shed was lost  muscle mass.</p>
<p>The statistics show both groups did indeed lose some  weight, but the first high protein group managed to lose even more  weight and retain muscle.</p>
<p>Dr. Donald Layman, the author of the  study, stated, &#8220;Both diets work because, when you restrict calories, you  lose weight. But the people on the higher-protein diet lost more  weight. There is an additive, interactive effect when a</p>
<p>protein  rich diet is combined with exercise. The two work together to correct  body composition; dieters lose more weight, and they lose fat, not  muscle.</p>

<p>The most common method used to pin down criminals in a court session  is through examining their DNA. So as to protect crime scenes from  outside interference, most forensic institutions keep these places off  limits to the general public. Criminals could have left finger prints,  blood samples, sweat droplets or hair strands as they struggled to flee  from the crimes scenes. These items are the ones that contain the DNA  that can be used to identify the criminals.</p>
<p>The best DNA tests can  be used to provide evidence in a court of law. A great example is blood  samples. The first thing that forensics do once blood is collected in a  crime scene is to identify the blood group. Thereafter, they will  identify the arrangement of DNA strands in blood components like  mitochondria. The arrangement of DNA in each individual is different. In  case they match perfectly with a suspected bad guy&#8217;s DNA arrangement  then that person is guilty. And if the blood group at the crime scene is  the same as this person&#8217;s, there is more reason to indict him/her. The  same process is used in other samples such a hair and sweat. To be more  accurate in their results researchers use at least 13 strands in  someone&#8217;s DNA for testing.</p>

<p>Mitochondrial DNA Testing was not even a thought until 1963 when it  was discovered to have it&#8217;s own separate DNA. Mitochondrial DNA (mtDNA)  is passed down maternally. Your mother passes mitochondria to both her  sons and daughters, though the sons do not pass it to their children.  Mitochondrial DNA gets passed from generation to generation maternally,  and the mtDNA does not change with slight exceptions.</p>
<p>Tracing your  ancestry has never had more solid proof then DNA testing, it is  virtually almost completely accurate. A book called &#8220;The Seven Daughters  of Eve&#8221; by Brian Sykes explains that we are all related to one common  ancestor he named Mitochondrial Eve. Throughout time these &#8220;7 daughters  of Eve&#8221; migrated into Europe (whom are all connected to Mitochondrial  Eve through there DNA) and many of us are descended from them. There are  around 30 other &#8220;daughters in the world, these seven daughters were  spread throughout Europe.</p>
<p>Researchers created a grouping system  called Haplogroups. These groups were made to show where these certain  groups of DNA migrated. Haplogroup H is the most common in Europe, this  &#8220;daughter is given the name &#8220;Helena&#8221;. About half of all Europeans are  descended from her. This Haplogroup originated in Southwest Asia Middle  East about 25,000 to 30,000 years ago.</p>

<p>The  order or specific sequences of the bases of the gene, provide the code  for making a specific protein. Proteins are composed of amino acids, and  each gene is responsible for producing a specific protein, which carry  out the life supporting work of living organisms.</p>
<p>There are several types of proteins with different functions that are central in the activities necessary for life.</p>
<p>Lab  techniques developed in recent years now make it possible to go beyond  looking at whole chromosomes or measuring the proteins of specific genes  and several types of tests can disclose details of the genes DNA.</p>
