Dec. 22, 2017 – For those with renal failure, the process of kidney dialysis is all too familiar: Three times a week, for a cumulative 12 hours, you sit in a chair and a machine does what your bean-shaped organs can’t — in this case, filter toxins out of your blood.

Although the treatment is an integral part of keeping those with advanced renal failure alive, patients’ quality of life leaves something to be desired. Dialysis of any kind is an unavoidable time sink, and one whose filtration systems have seen little change over the years.

But what if there was a way to make the process faster — and cheaper?

Saeed Moghaddam, Ph.D., an associate professor in UF’s Department of Mechanical and Aerospace Engineering, is developing a membrane with the potential to revolutionize this process by maximizing its efficiency.

In order for dialysis to eventually become something that patients may undergo from the comfort of their own home, Moghaddam identified two things that needed to change: the configuration of the membrane, and the material from which it was made.

“This membrane is based on simplicity,” Moghaddam says. “It is based on something that uses forces of physics, rather than brute force. I made it simple through better understanding and utilizing molecular and nanomaterials self-assembly processes.”

Moghaddam believes that new generation functional life support systems can be developed by advancing the science of materials interactions at both the molecular and nanoscale. In this case, the membrane is composed of two sheets of graphene, one of the thinnest possible existing molecular sieves. The two layers stagger the man-made pores in each sheet, lowering the chances of critical substances passing through.

As barbers joke that the secret to a good haircut is knowing what to leave behind, the same holds true for biomedical filtration.

“The purpose is to be selective,” Moghaddam says. “You cannot keep losing red blood cells, albumin, or other valuable proteins through defects inherent to typical micro-and nanomanufacturing methods.”

When it comes to filtration, chemically modified graphene sheets are unparalleled in their adsorptive nature and selective permeability. Daniel Ornt, a clinical nephrologist and the Vice President of Health Sciences at the Rochester Institute of Technology, made sure that the project’s development and testing mimicked a clinical environment as closely as possible. For example, considering how patient experiences might be affected.

“The duration of treatment and size of the filter could be dramatically improved, cutting down on time spent at a clinic,” Ornt says. “It would also lend the dialysis process more speed and selectivity.”

But not only does the engineering behind the membrane speed up its intended purpose, it takes into account the production value and bigger goal.

“Both of us have the philosophy of looking for scalable nanomanufacturing that can be applied to significant problems our country and society faces,” says Thomas Gaborski, Ph.D. the other PI on the NIH-backed project and associate professor of biomedical engineering at the Rochester Institute for Technology.

“We don’t just want to make a new nanomaterial discovery in our lab,” Gaborski says. “We want to translate this into a technology that can be useful beyond just the limited confines of our own research laboratories.”

Accordingly, graphene is a material that is comparatively quick and inexpensive to mass-produce, making this developing membrane a potentially uniquely positioned solution for the thousands on waiting lists for organ transplants.

“A liver or kidney transplant is always the best solution, but the supply is woefully inadequate around the world,” Gaborski says. “Technologies like this are necessary to help patients until that supply becomes available.”

Gaborski and Moghaddam’s project has received support from NIH and is just one of many recent interdisciplinary collaborations involving the University of Florida. Increasingly, research efforts at the university seek to explore the intersection between medicine and engineering, with innovative and promising results.

“By translating ideas from one discipline to another, I believe that we can have a really positive impact,” says David Hahn, chair of the Department of Mechanical and Aerospace Engineering. “It’s this interdisciplinary approach that we’re hoping to ingrain in our students — the idea that problem-solving can be much more effective, and more efficient if you consider a diversity in approaches.”

Duchenne muscular dystrophy is caused by a faulty gene that leads to progressive muscle weakness

May 14, 3017 – Researchers at the National Institutes of Health’s National Center for Advancing Translational Sciences (NCATS) and the University of Nevada, Reno School of Medicine (UNR Med) have demonstrated that a drug originally targeted unsuccessfully to treat cancer may have new life as a potential treatment for Duchenne muscular dystrophy (DMD).

The candidate drug, SU9516, represents a different kind of approach for treating DMD, a degenerative muscle disease that usually begins in childhood and has no known cure. It is caused by a faulty gene that leads to progressive muscle weakness, with death often occurring around age 25. Rather than trying to fix or replace the broken gene, SU9516 ramps up the muscle repair process, helping reinforce muscle structure.

NCATS Chemical Genomics Center Acting Branch Chief Juan Marugan, Ph.D., and UNR Med Professor of Pharmacology Dean Burkin, Ph.D., led a team that screened more than 350,000 compounds to find SU9516, which had been previously developed as a treatment for leukemia. The research demonstrated that this compound improved muscle function in both laboratory and animal DMD models. The results, published recently in Molecular Therapy, may provide a promising approach against the disorder and other muscle-wasting conditions.

Those with DMD lack dystrophin, a protein akin to a molecular shock-absorber that helps keep muscle cells intact. Without dystrophin, muscles are fragile and easily injured. Individuals lose muscle strength and the ability to repair damaged muscle tissue. Most die from heart or respiratory problems.

“Our findings open the door to develop new drug treatments for DMD,” Marugan said.

In earlier research, Burkin, who is senior author of the current study, and his co-workers showed that boosting the levels of a cell structural protein, α7β1 integrin, in affected muscle cells could alleviate DMD symptoms in a mouse model. In addition, increased amounts of the protein slowed the disease’s progress.

Burkin and his UNR Med colleagues collaborated with NCATS researchers, including co-team leaders Marc Ferrer, Ph.D., and Noel Southall, Ph.D., to screen a large collection of compounds for molecules that could increase α7β1 integrin production in mouse muscle cells grown in the laboratory. The screen revealed that SU9516 raised integrin production and promoted the formation of muscle cells and fibers from DMD muscle stem cells, another important indication of its potential as a drug.

In a series of pre-clinical experiments, the researchers showed that SU9516 increased the production of α7β1 integrin in human and mouse DMD muscle cells. Subsequent tests found SU9516 improved muscle function and slowed indicators of disease progression.

Burkin suggests that such a drug could be used alone, or in combination, with other therapies yet to be developed. There might be wide ranging applications to other muscle-damaging conditions, like cachexia, a wasting syndrome characterized by weight loss and muscle atrophy that is often seen in the late stages of cancers, and the effects of aging and injury, he noted.

“Integrin stabilizes muscle structure, and helps stimulate muscle repair and regeneration,” Burkin said. “If we can artificially increase its production with drugs, we think it can help protect muscle cells from damage.”

The NCATS-UNR Med team plans to work with medicinal chemists to make the molecule more specific for DMD, while also removing the toxic anticancer components, creating a safer version with a goal of future testing in patients.

Epidemiological evidence from human case studies showed that within weeks or even days of getting the Tdap shot, children developed infantile spasms and other common symptoms of microcephaly directly related to the vaccine.

Earlier research cited in this same government report from 1987 pinpoint what scientists described as infantile spasms immediately following pertussis immunization. Six children evaluated as part of this earlier research all developed seizures and other problems in conjunction with getting the pertussis vaccination, which the authors described as being caused by one of the following:

• A direct neurotoxic effect (from the vaccine)
• An immediate immune reaction (from the vaccine)
• A delayed cellular hypersensitivity reaction (from the vaccine)
• A vaccine-induced activation of a latent neurotropic virus infection (obviously from the vaccine)

Each of these effects is directly related to the vaccine itself, illustrating an abysmal safety record that isn’t all puppies and roses as popular opinion often dictates. Numerous studies published between the 1950s and the 1970s also cited in the government report demonstrate clear cause-and-effect damage resulting from both the pertussis and Tdap vaccines, with only a very small percentage of cases of damage possibly resulting from prenatal factors.

“Among symptomatic cases, presumed causes are frequently grouped according to the timing of the suspected insult as occurring pre-, peri-, or postnatally,” the study explains. “Prenatal factors are thought to account for 20 to 30 percent of cases. Are microcephaly outbreaks caused by Zika as the media claims, or by vaccines and other sources of chemical exposure?

This thorough review of vaccine safety by the U.S. government isn’t some conspiracy theory; it’s a repository of evidence compiled from all the available evidence at the time as to post-vaccine outcomes of children, and the results were disastrous. Many children appear to suffer major neurological abnormalities following vaccination with pertussis and/or Dtap, but you’d be hard-pressed to hear a peep about this today from the likes of the U.S. Centers for Disease Control and Prevention (CDC).

The paper is even so bold as to proclaim that the various case reports of children who develop infantile seizures following vaccination for pertussis specifically “prompt concern” as to the vaccine’s safety. The many case reports, case series, and controlled observational epidemiological studies summarized in the paper all point to a direct relationship between the Dtap vaccine and infantile seizures.

Interestingly enough, the microcephalic impact of both the Dtap and pertussis vaccines is far more evident than with the Zika virus, which the media has been going nuts over as the cause of various observed microcephaly outbreaks in recent months.
