Characteristics of Corning Centrifuge Tubes .

The following information is provided to serve as a general guideline for determining suitability of Corning centrifuge tubes for your applications. In addition, Corning recommends following the procedures outlined by the centrifuge manufacturer, as well as conducting a trial run to determine proper conditions before beginning any critical applications.
Corning centrifuge tubes are tested for leakage. They should not break or leak if used in a properly balanced rotor with suitable carriers, holders, and adapters that fully support the tubes when run in accordance with the guidelines in this section. These tubes are intended for one-time use only; reuse is not recommended as breakage or leakage may occur.
The recommended working temperature range for Corning centrifuge tubes is 0°C to 40°C. The suitability of these tubes for storage below 0°C depends on both the solution and the storage conditions. In general, the polypropylene and PET tubes are more resistant to stress at low temperatures than polystyrene. Suggestions for Safe Centrifugation

When centrifuging pathogenic organisms, specimens known or suspected of being infectious, or any other potentially infectious or hazardous materials, approved safety containment systems should be used. Contact your centrifuge manufacturer for appropriate accessories or recommendations.
Proper balancing and distribution of the load in a centrifuge is critical for optimum performance and to prevent damage to the tubes or centrifuge. Opposing buckets or loads should always be balanced within the range specified by the manufacturer. Tubes should always be distributed in the buckets with respect to the center of rotation as well as the pivotal axis of the bucket. Failure to do this may prevent the bucket from achieving a horizontal position during the centrifugation run. Uneven separations or tube failure may result.
These centrifuge tubes are intended for use by persons knowledgeable in safe laboratory practices. Failure can result from surface damage, exceeding the specified RCF values, using unsuitable support systems, improper temperatures, or incompatible chemicals.

Filtration centrifuges (filter centrifuges or screen centrifuges)

The used filter centrifuge has a perforated drum (similar to a spin dryer) which is covered on the inside by a suitable (harmonized to the respective product) filter insert.

These filter inserts in a centrifuge can, for example, be:

Filter cloth (peeler centrifuge, inverting filter centrifuge, pendulum centrifuge)
Wire mesh of perforated plate (worm screen centrifuge)
Slot screen (pusher centrifuge, swarf centrifuge)

In the centrifuges, the suspension flows from the inside to the outside through the filter insert and the perforated wall of the drum, whereby the solid material is restrained by the filter insert. Depending on the type of centrifuge, the restrained solid material can be removed from the centrifuge as follows:

manually (tubular centrifuge, certain types of pendulum centrifuges)
periodically: by a peeling blade being periodically driven into the drum (peeler centrifuges)
periodically: by the filter cloth periodically being inverted (inverting filter centrifuge)

Keep all reagents to room temperature(15-30℃) prior to use. The test should be carried out at room temperature and the operation procedure is described below:
1.Clean your mouth 30 minutes before collecting the salivaPlace the tip oftongue against the upper or lower tooth root to enrich saliva, and then swallow saliva to the throat without ingesting saliva to esophagus, followed by returning saliva to mouth repeat for 5 times.
2. Deliver saliva into the collection cup.
3.Insert the sample diluent tube into the pipe rackmake sure that the tube is standing firm and reaches the bottom of the pipe rack.
4.Open the purple cap of the sample diluent tubeadd 12 drops(about 180ul) of collected saliva into the sample diluent tube which contains 0.3 mL ofthe diluent buffer.
5.Screw cap onto the collection tube,and invert gently 5 times to mix the saliva and sample diluent buffer thoroughly.
6.Open the small cap on the top of the sample dilution tube.Add 3-4 drops(~100uL) of the sample diluent buffer immediately to the sample well,allow the strip to develop for 10~15 minutes at room temperature. A visible band can be read by naked eyes. The INDICAID COVID-19 Antigen At-Home test should be carried out at room temperature

Explanation of the Test

COVID-19 (short for “Coronavirus disease 2019”) is a disease first recognized in 2019 that is caused by type of novel coronavirus called SARS-CoV-2. Due to its rapid spread, the World Health Organization (WHO) recognized the disease as a global pandemic on March 11, 2020. Many individuals with confirmed COVID-19 have developed fever and/or symptoms of acute respiratory illness (e.g., cough, dyspnea), although some individuals experience only mild symptoms or no symptoms at all.

The virus is spread primarily from person to person through respiratory particles, even by individuals without symptoms. The INDICAID COVID-19 Antigen At-Home Test is a rapid qualitative
immunochromatographic assay for the determination of the presence of SARSCoV-2 antigens in anterior nasal swab specimens. Each INDICAID COVID-19 Antigen At-Home Test is single-use and can analyze one anterior nasal (nares) swab sample. The total time required to perform one test is approximately 20
minutes from clinical specimen collection to result.

SARS-CoV-2-specific antibodies and a control antibody are immobilized onto a nitrocellulose membrane support as two distinct lines. The test line (T) region contains monoclonal anti-SARS-CoV-2 antibodies and the control line (C) region contains polyclonal control antibodies. Polyclonal and monoclonal anti-SARS-CoV-2 antibodies conjugated with red-colored latex microspheres are used to detect the
SARS-CoV-2 antigen.

During the test, the swab containing patient sample is placed and mixed in a Buffer Solution Vial. That Buffer Solution is then applied to the sample well of the test device. If SARS-CoV-2 antigen is present, it will bind to the antibody-latex microsphere conjugate forming an immunocomplex. The immunocomplex will then travel across the strip via capillary action towards the test line. The immunocomplex will then bind to the anti-SARS-CoV-2 antibodies at the test line (T), forming a visible red-colored line to indicate detection of antigens. If SARS-CoV2 antigens are not detected in the sample, no color will appear at the test line (T).

Test results are interpreted visually at 20 minutes after the sample has been properly applied to the test according to the instructions. Results should not be read after 25 minutes.
The control (C) line is used for procedural control and should appear regardless of the test result. The appearance of the control line (C) serves to ensure the test is performing properly and the test result is valid. The INDICAID COVID-19 Antigen At-Home Test is validated for use from direct
specimens testing without transport media.

Warnings and Precautions

• For in vitro diagnostic use only.
• This product has not been FDA cleared or approved, but has been authorized by FDA under an Emergency Use Authorization (EUA).
• This product is only authorized for the detection of proteins from SARS-CoV2, not for any other viruses or pathogens.
• The emergency use of this product is only authorized for the duration of the declaration that circumstances exist justifying the authorization of emergency use of in vitro diagnostics for detection and/or diagnosis of COVID-19 under Section 564(b)(1) of the Act, 21 U.S.C. § 360bbb-3(b)(1), unless
the declaration is terminated or authorization is revoked sooner.

Read the Instructions for Use carefully before performing test. Failure to
follow directions may produce inaccurate test results.

• Wear a facemask when collecting specimen from another individual.
• Do not use this test kit beyond the expiration date printed on the outside of
the box.
• Do not use if any of the test kit contents or packaging is damaged.
• Do not use the test on children under 2 years of age.
• Use only the contents provided in the test kit.
• All test components are single use. Do not re-use.
• Wash hands thoroughly for at least 20 seconds before and after using the
test.
• Do not open the kit contents until ready for use. Use within 2 hours of
opening the Test Device pouch.
• Leave the swab inside its packaging until instructed to swab the nose. Keep
the swab clean. Do not allow anything to touch the soft tip of the swab until
instructed to swab the nose.
• When collecting a sample, use only the nasal swab provided in the kit.
• Perform the test as soon as possible after swabbing both nostrils, and within
30 minutes after adding the swab to the vial.
• This test is read visually. User with impaired vision or color impaired vision
may not be able to read the test.
• Do not interpret the test result before 20 minutes or after 25 minutes,
following application of the sample to the Test Device.
• False negative results may occur if insufficient Buffer Solution is applied to
the Test Device (e.g. less than 3 drops).

Storage and Stability

• The INDICAID COVID-19 Rapid Antigen At-Home Test should be stored in a
cool, dry place between 2-30°C (35.6-86°F). Do not freeze. Avoid direct sunlight.
• Kit components in the INDICAID COVID-19 Rapid Antigen At-Home Test are stable until the expiration date printed on the label.

SARS-CoV-2: An overview of virus structure, transmission and detection

Severe Acute Respiratory Syndrome Coronavirus 2 (SARS-CoV-2) is an enveloped, single-stranded RNA virus of the family Coronaviridae. Coronaviruses share structural similarities and are composed of 16 nonstructural proteins and 4 structural proteins: spike (S), envelope (E), membrane (M), and nucleocapsid (N). Coronaviruses cause diseases with symptoms ranging from those of a mild common cold to more severe ones such as Coronavirus Disease 2019 (COVID-19) caused by SARS-CoV-2 ^1,2.

SARS-CoV-2 is transmitted from person-to-person primarily via respiratory droplets, while indirect transmission through contaminated surfaces is also possible^3-6. The virus accesses host cells via the angiotensin-converting enzyme 2 (ACE2) receptor, which is most abundant in the lungs^7,8.

The incubation period for COVID-19 ranges from 2 – 14 days following exposure, with most cases showing symptoms approximately 4 – 5 days after exposure^3,9,10. The spectrum of symptomatic infection ranges from mild (fever, cough, fatigue, loss of smell and taste, shortness of breath) to critical^11,12. While most symptomatic cases are not severe, severe illness occurs predominantly in adults with advanced age or underlying medical comorbidities and requires intensive care. Acute respiratory distress syndrome (ARDS) is a major complication in patients with severe disease. Critical cases are characterized by e.g., respiratory failure, shock and/or multiple organ dysfunction, or failure.

The SARS-CoV-2 Rapid Antigen Test is a lateral fl ow rapid chromatographic immunoassay for the
qualitative detection of nucleocapsid antigen to SARS-CoV-2 present in human nasal samples. This test is intended for use as an aid in detection of SARS-CoV-2 infection in individuals suspected of COVID-19 with clinical symptoms onset within 5 days. Results are for the identification of SARS-CoV-2 nucleocapsid antigen. Antigen is generally detectable in human nasal swab samples during the acute phase of infection. Positive results indicate the presence of viral antigens, but clinical correlation with patient history and other diagnostic information is necessary to determine infection status. Positive results do not rule out bacterial infection or coinfection with other viruses. The agent detected may not be the definite cause of disease.
Negative results should be treated as presumptive, and do not rule out SARS-CoV-2 infection and
should not be used as the sole basis for treatment or patient management decisions, including
infection control decisions. Negative results should be considered in the context of a patient’s recentexposures, history and the presence of clinical signs and symptoms consistent with COVID-19, and confirmed with a molecular assay, if necessary, for patient management. The SARS-CoV-2 Rapid Antigen Test is intended for use in laboratory or POC settings by healthcare professionals, or
self-collection under the supervision of a healthcare worker.

INTRODUCTION

Coronaviruses can cause a variety of acute and chronic diseases. Common signs of a person infected
with a coronavirus include respiratory symptoms, fever, cough, shortness of breath, and dyspnea. In
more severe cases, infection can cause pneumonia, severe acute respiratory syndrome, kidney failure,

PRINCIPLE OF THE TEST

The SARS-CoV-2 Rapid Antigen Test has two pre-coated lines: A “C” Control line and a “T” Test line
on the surface of the nitrocellulose membrane. Both the control line and test line in the result
window are not visible before applying any samples. Mouse monoclonal anti-SARS-CoV-2 antibody is coated on the test line region and mouse monoclonal anti-Chicken IgY antibody is coated on the control line region. Mouse monoclonal anti-SARS-CoV-2 antibody conjugated with color particles are used as detectors for the SARS-CoV-2 antigen device.

During the test, the SARS-CoV-2 antigen in the sample interacts with monoclonal anti-SARS-CoV-2 antibody conjugated with color particles making an antigen-antibody color particle complex. This complex migrates on the membrane via capillary action to the test line, where it is captured by the mouse monoclonal antiSARS-CoV-2 antibody. A colored test line becomes visible in the result window if SARS-CoV-2 antigens are present in the sample.

Vaccination is one of the most important and economic tools to protect the community from infectious diseases. There are about 60% of the population were fully vaccinated around the world, however, as the virus mutates continuously, the increasingly strong transmission and immune escape capabilities of the mutant strains have brought great challenges to the prevention and control of the epidemic. The emergence of many breakthrough infection cases represents that the “immune barrier” created by vaccination against the wild-type strain of COVID-19 may be flawed. Therefore we INDICAID urgently need next-generation vaccines that still have high protective efficacy against mutant strains.

As the world experiences a new wave of outbreaks due to the emergence of the Omicron variant, vaccine manufacturers have already stepped up for the development of vaccines against new variants. Earlier, Moderna and Pfizer had announced to start the research on candidate vaccines against Delta and Omicron variants. In China, the China Center for Disease Control and Prevention, Sinovac and Sinopharm Group received the Omicron variant virus isolated by HKU and began their work. In addition, CanSino Bio said they were also working on the development of a new vaccine against the Omicron variant.
In order to support and regulate the development of COVID-19 vaccines, the Food and Drug Administration (FDA) issued the “Development and Licensure of Vaccines to Prevent COVID-19” to determine key indicators for monitoring the whole process of vaccine development.

This test is also intended for non-prescription home use with adult-collected anterior nasal (nares) swab samples from individual aged 2 years or older with symptoms of COVID-19 within the first 6 days of symptom onset. This test is also authorized for non-prescription home use with self-collected
anterior nasal (nares) swab samples from individuals aged 14 years or older, or adult-collected anterior nasal (nares) swab samples from individuals aged 2 years or older, with or without symptoms or other epidemiological reasons to suspect COVID-19 when tested twice over three days with at least 24 hours (and not more than 48 hours) between tests.

The INDICAID COVID-19 Antigen At-Home Test does not differentiate between SARS-CoV and SARS-CoV-2.

Results are for the identification of SARS-CoV-2 nucleocapsid protein antigen. Antigen is generally detectable in anterior nasal (nares) samples during the acute phase of infection. Positive results indicate the presence of viral antigens, but clinical correlation with past medical history and other diagnostic information is necessary to determine infection status. Positive results do not rule out bacterial infection or co-infection with other viruses and the agent detected may not be the definite cause of disease. Individuals who test positive with the INDICAID COVID19 Antigen At-Home Test should self-isolate and seek follow-up care with their physician or healthcare provider as additional testing may be necessary.
Negative results should be treated as presumptive and confirmation with a molecular assay, if necessary, for patient management, may be performed. Negative results do not rule out SARS-CoV-2 infection and should not be used as the sole basis for treatment or patient management decisions, including infection control decisions. Negative results should be considered in the context of a patient’s recent exposures, history, and the presence of clinical signs and symptoms consistent with COVID-19.

Stability

Assay Principle

This kit is based on sandwich enzyme-linked immuno-sorbent assay technology. An antibody is pre-coated onto a 96-well plate. Standards, test samples, and biotin-conjugated reagent are added to the wells and incubated. The HRP-conjugated reagent is then added, and the whole plate is incubated. Unbound conjugates are removed using wash buffer at each stage. TMB substrate is used to quantify the HRP enzymatic reaction. After TMB substrate is added, only wells that contain sufficient SPON2 will produce a blue coloured product, which then changes to yellow after adding the acidic stop solution. The intensity of the yellow colour is proportional to the SPON2 amount bound on the plate. The Optical Density (OD) is measured spectrophotometrically at 450 nm in a microplate reader, from which the concentration of SPON2 can be calculated.

Assay Procedure

1) Set standard, test samples and control wells.
2) Aliquot 100 µl of diluted standard into the standard wells.

5) Aliquot 100 µl of Detection Reagent A to each well. Incubate for 1 hr at 37 °C.
6) Wash 3 times.
7) Aliquot 100 µl of Detection Reagent B to each well. Incubate for 30 mins at 37 °C.
8) Wash 5 times.
9) Aliquot 90 µl of TMB Substrate to each well. Incubate for 10-20 mins at 37 °C.
10) Aliquot 50 µl of Stop Solution.
11) Measure the OD at 450 nm.

Protocol

Equilibrate the kit components and samples to room temperature (18 – 25 °C) before use. It is recommended to plot a standard curve for each test.
Set standard, test sample and control (zero) wells on the pre-coated plate respectively, and then, record their positions. It is recommended to measure each standard and sample at least in duplicate.
Add 100 µL of each standard, control and sample into the appropriate wells. Seal the plate with a cover and incubate for 1 h at 37°C.
Remove the cover and discard the liquid.
Add 100 µl of the detection Reagent A working solution to each well. Seal the plate with a cover and incubate for 1 h at 37°C.
Remove the cover and discard the solution. Wash the plate 3 times with 1X Wash Buffer.
Add 100 µL of Detection Reagent B working solution into each well, seal and incubate at 37°C for 30 min.
Discard the solution and wash the plate 5 times with wash buffer as explained in previous step.
Aliquot 90 µl of TMB Substrate into each well. Seal the plate with a cover and incubate at 37°C for 10-20 min. Avoid exposure to light. The incubation time is for reference use only, the optimal time should be determined by end user. Do not exceed 30 min.
Add 50 µL of Stop Solution to each well. Read at 450 nm immediately.

Quality

Function test: The preparation is used as template in a PCR with the Expand PCR System and appropriate primers from the human tPA Control Primer Set. Amplification products up to 27kb long are obtained.
Absence of contaminating organisms: The serum used for this preparation was tested for HBs antigen and the presence of antibodies to HIV-1, HIV-2, HCV. All tests were negative.

What is Genomic DNA?

Genomic DNA constitutes the total genetic information of an organism. The genomes of almost all organisms are DNA, the only exceptions being some viruses that have RNA genomes. Genomic DNA molecules are generally large, and in most organisms are organized into DNA–protein complexes called chromosomes.

The size, number of chromosomes, and nature of genomic DNA varies between different organisms (see table Sizes and molecular weights of various genomic DNAs). Viral DNA genomes are relatively small and can be single- or double-stranded, linear, or circular. All other organisms have double-stranded DNA genomes. Bacteria have a single, circular chromosome.

In eukaryotes, most genomic DNA is located within the nucleus (nuclear DNA) as multiple linear chromosomes of different sizes. Eukaryotic cells additionally contain genomic DNA in the mitochondria and, in plants and lower eukaryotes, the chloroplasts. This DNA is usually a circular molecule and is present as multiple copies within these organelles.

Chemical and Synthetic Biology Approaches To Understand Cellular Functions

Preparation of genomic DNA

gDNA must be appropriately fragmented before being transformed into yeast spheroplasts. Previous studies have demonstrated that homologous recombination combining two linear pieces of DNA into a yeast artificial chromosome is much more efficient if the homologous arm sequences are closer to the ends of the targeted DNA fragment (Noskov et al., 2003).

Ideally, site specific double-stand breaks (DSBs) can be introduced directly upstream and downstream of BGCs of interest by restriction enzyme digestion, and homology arm sequences should be designed as close to these restriction sites as possible. However, this approach can be limited due to the lack of appropriate restriction sites directly beyond and not within BGCs of interest. Alternatively, gDNA can be randomly fragmented by shearing with a needle or pipette tip.

Recently, Dr. Natalay Kouprina and colleagues have successfully introduced DSBs to the ends of targeted gDNA fragments using the CRISPR-Cas9 endonuclease in vitro, resulting in high efficiency DNA capture, up to 32% for large fragments isolated from complex gDNA (Lee, Larionov, & Kouprina, 2015). However, in vitro CRISPR/Cas9-mediated DNA digestion necessitates several troubleshooting steps and thus may not be an attractive approach if commercial restriction enzymes can be used.

The quality of the gDNA also greatly affects recombination efficiency. For isolation of gDNA from a single organism in pure culture, standard gDNA isolation procedures or commercial isolation kits are generally adequate, but gDNA should be checked by Nanodrop, Qubit, and agarose gel to confirm integrity. Finally, if the target of interest is low in quantity (such as from metagenomic DNA), PCR and/or genomic library preparation are encouraged for enrichment.

Genome Sequence Databases: Genomic, Construction of Libraries.

Genomic DNA must be isolated from proteins and other cellular debris prior to any enzymatic or mechanical manipulation. Bacterial cells are lysed, typically through exposure to surfactants, such as sodium dodocyl sulfate or Tween-20, or treatment with lysozyme to digest the polysaccharide component of cellular membranes and proteinase K for protease digestion. DNase-free RNase may be added to the lysis step to minimize RNA contamination.
Genomic DNA can be purified from cell lysate using a phenol–chloroform extraction followed by an ethanol precipitation or commercially available silica columns. Commercially available kits for genomic DNA isolation are often desirable in that they avoid the use of phenol and chloroform, which are toxic and may interfere with downstream enzymatic reactions. Many commercially available kits avoid phenol by using buffers containing the chaotropic agent guanidine hydrochloride to aid in cell lysis and to effectively denature proteins. Purified genomic DNA should be maintained in a nuclease-free Tris buffer or in nuclease-free water.
Nuclease contamination is a frequent concern associated with genomic DNA isolation. Nuclease activity will degrade DNA and can be easily mistaken for a restriction enzyme digestion or the result of mechanical shearing. Nuclease contamination may be detected by incubating an aliquot of purified DNA at 37 °C for 18 h and then visualizing the DNA on an agarose gel.
A control aliquot of DNA that had been stored frozen should be used for comparison. Following electrophoresis, if the incubated aliquot appears to have degraded, nucleases may be contaminating the genomic DNA sample. Additionally, the DNaseAlert kit available from Ambion (Austin, TX) can be used to detect DNase contamination in a sample.
This kit detects DNase in a sample through the use of modified oligonucleotides that fluoresce upon cleavage by nucleases that may be within the sample. If nuclease activity is detected, the DNA sample can be exposed to high concentrations of guanidine hydrochloride (6–8 M) for 30 min or be subjected to an additional phenol–chloroform extraction for more thorough deproteinization of the sample. An ethanol precipitation should follow the additional deproteinization step to remove the phenol, chloroform, or chaotropic salts.

2) Take 10ul of serum, plasma, or whole blood to the dark score line of a
capillary tube.
3) Slowly add 10 ul of serum, plasma, or whole blood to the sample well
with capillary tube with a score line for volume of 10ul and then add 2
drops (approximately 60ul) with bottle containing diluent buffer. If the
migration is not appeared whitin 1 minute, add one more buffer to sample
well. 4) As the test result, you can see the purple band in the result window of the
kit.
5) Interpret test results at 20 minutes.
6) Please do not read after 20 minutes.

Interpretation of the Test

1) A color band which would be appeared in the left section of the result
window shows that the test is works properly, and it called is the Control
line (C).
2) The right section of the result window called test band indicates the test
results. If another color band appears on the test band. (T).
Negative : The presence of only one purple color band within the result
window indicates a negative result.

Positive : The presence of two color bands (“T” band and “C” band) within
the result window, no matter which band appears first, means
positive.

Invalid : If the purple color band is not visible within the result window
after performing the test, the result is considered invalid. The
directions may not have been followed correctly or the test may
have deteriorated. It is recommended to take the test again with
same specimen.

Limitations of the Test

1) The Anigen Rapid C. Brucella Ab Test Kit will only indicate the antibody
presence against Brucella canis in the specimen.
2) As with all diagnostic tests, all results must be interpreted together with
other clinical information available to the veterinarian.
3) If the test result is negative and clinical symptom is persist, additional
testing using other clinical methods is recommended. A negative result
does not preclude the possibility of canine brucellosis.
9. Expected Values
The Anigen Rapid C. Brucella Ab Test Kit has been compared with 2-
mercapto-ethanol Rapid Slide Agglutination Test. The overall accuracy is
greater or equal to 90.0%.

Background

Bovine brucellosis is commonly caused by Brucella abortus and less frequently by B. melitensis, and rarely by B. suis. Infection is widespread globally. Humans may be infected by contact with animals or animal products contaminated with these bacteria. Available serological tests include the Rose Bengal, ELISA, complement fixation test and tube agglutination test. However, these tests do not provide rapid diagnosis and each requires specialised laboratories and equipment. The new Anigen immunochromatographic rapid assay was developed to provide accurate, rapid and easy testing of B. brucellosis.

The rapid slide agglutination test (RSAT) for the diagnosis of Brucella canis was described by George and Carmichael (Proc. Council Res. Workers in Animal Disease, November 1973). The basis of the Genprice Gentaur test is direct agglutination of the killed stained antigen by Brucella canis antibodies.

GENERAL INFORMATION

Canine brucellosis is a chronic infection with Brucella canis that causes generalized lymphadenitis and mild to severe reproductive symptoms. Prostatitis, epididymitis, scrotal dermatitis, testicular atrophy and impotence can be present in the male; abortion typically in the final two weeks of gestation, resorption, infertility, and vaginal discharge can be present in the female.
Brucella canis is infectious to humans. Caution should be exercised when handling serums to be tested.
The test has been shown to presumptively diagnose infection with Brucella canis by detecting specific antibody that is formed 1 to 4 weeks after infection. The basis for serodiagnosis of canine brucellosis is the 2-Mercaptoethanol Tube Agglutionation Test (2ME-TAT) and the Rapid Slide Agglutination Test (RSAT).
The 2ME-TAT and RSAT have demonstrated excellent correlation in experimentally infected dogs. In field situations, it has been recognized that an occasional healthy dog, culturally negative for
Brucella canis will react positively in the RSAT, but not in the 2ME-TAT. The 2-Mercaptoethanol – Rapid Slide Agglutination Test (2ME-RSAT) has been developed in an attempt to eliminate discrepancies between the RSAT and the 2-ME-TAT.
Certain non-specific agglutinins, reported to occur in the sera of normal dogs, are removed from canine sera when 2-mercaptoethanol is employed in the 2ME-TAT. Because of this occurrence, 2-
mercaptoethanol is employed in the modified Rapid Slide Agglutination Test procedure.

INDICATIONS

The reagents are used to presumptively diagnose infection with Brucella canis in dogs. Positive results
require additional confirmation.

PRECAUTIONS

Antigen and accompanying antiserum have been standardized and should be used together. Store
components at 2-7C (35-45F). Do not allow reagents to stand at room temperature for excessive
periods of time. Do not freeze.

SAMPLE INFORMATION
Serum is required. Test may not be used with whole blood or plasma.

Procedure for Rapid Slide Agglutination Test (RSAT)
1. ENSURE REAGENTS AND SAMPLE ARE AT ROOM TEMPERATURE (70-78F; 21-25C) prior
to testing.
2. Cut card on dotted line. Two circles are used to perform each test.
3. Place one drop of Reagent A, Positive Control (Red Cap), in one circle of the supplied card.
4. Use a disposable plastic pipette to place one drop of test serum in another circle. Each test kit
contains disposable pipettes to prevent serum cross contamination. Do not dispose of pipette until
the 2ME-RSAT is completed.
5. Thoroughly mix Reagent B, Antigen (White Cap), by vigorously shaking the vial. Add one drop
of the Reagent B, Antigen, close to each serum drop, being careful not to touch the serum with
the dropper tip.
6. Mix each antigen-serum combination with a separate end of a stir stick, spreading to fill the circular
area. Do not allow the positive control test to touch the unknown serum sample.
7. Gently rock the card for 10-15 seconds. Place on a flat surface and observe for agglutination for
no longer than two minutes. If the serum is negative (absence of agglutination), the animal is
considered not to be infected with B. canis. If the card tests positive, perform the 2ME-RSAT.

Procedure For 2-Mercaptoethanol-Rapid Slide Agglutination Test (2ME-RSAT)

1. Add 2 drops of Reagent C, 2-Mercaptoethanol (Yellow Cap) to a tube containing 2 drops of the
serum to be tested, and mix well.
2. Place 1 drop of this mixture on a new reaction circle.
3. Add 1 drop of Reagent B, Antigen as above to the serum solution and mix with stir stick as
described above.
4. Gently rock the card for 10-15 seconds. Place on a flat surface and observe for agglutination for
no longer than 2 minutes. When the RSAT-positive sample also tests positive by 2ME-RSAT, the
animal is presumptively diagnosed as being infected with B. canis. Blood should be subjected to
cultural examination for B. canis. When the RSAT-positive samples tests negative by 2ME-RSAT,
the animal may be in the early stage of B. canis infection, or alternatively, its serum may contain
non-specific agglutinins to B. canis. To distinguish between these two conditions, a second serum
sample should be collected in approximately thirty days and retested by the 2ME-RSAT procedure.
Only if this sample tests positive should the animal be presumptively diagnosed as having B. canis
infection. Blood should then be subjected to cultural examination for B. canis.
Definitive diagnosis of canine brucellosis is based upon isolation of B. canis from the animal.

Background

Brucellosis, also known as undulant fever, Mediterranean fever is a zoonosis and the infection is almost invariably transmitted by direct or indirect contact with infected animals or their products. Expansion of animal industries and urbanization, and the lack of hygienic measures in animal husbandry and in food handling partly account for brucellosis remaining a public health hazard. Expansion of international travel which stimulates the taste for exotic dairy goods such as fresh cheeses which may be contaminated, and the importation of such foods into Brucella-free regions, also contribute to the ever-increasing concern over human brucellosis.

The Anigen Rapid B.Brucella Ab Test Kit is a chromatographic immunoassay for the qualitative detection of antibodies against Brucella abortus in bovine whole blood, plasma, serum or milk.
The Anigen Rapid B.Brucella Ab Test Kit has two letters which are test(T) line and control(C) line on the surface of device. The test line and control line in the result window are not visible before applying any samples. The control line is a reference line which indicates the Genprice Gentaur test is performing properly. The control line has to appear every time when the test has performed. If the antibodies against Brucella abortus are present in sample, a purple test line would appear in the result window.

Precautions

The test kit is for canine use only. Do not use for other animals.
The test device is sensitive to humidity as well as heat. Perform the test immediately after removing the test device from the foil pouch. Do not re-use the test components.
Apply the sample and assay diluent vertically.
Do not touch the membrane in the result window of test device.
Do not use the test kit beyond the stated expiration date marked on the
package label.
Do not use the test kit if the pouch is damaged or the seal is broken.
Do not mix components from different lot numbers because the components in this kit have been quality control tested as standard batch unit.
All samples should be handled as being potentially infectious. Wear protective gloves while handling samples. Wash hands thoroughly afterwards.
Decontaminate and dispose of all samples, reaction kits and potentially contaminatedmaterials safely in accordance with national and local regulations.

Limitations of the Test

Although the Anigen Rapid C.Brucella Ab Test kit is very accurate for detecting antibodies to Brucella canis, but a low incidence of false results can occur. Other clinical tests are required if questionable results are obtained. As with all diagnostic tests, a definitive clinical diagnosis should not be based on the results of a single test, but should only be made by the veterinarian after all clinical and laboratory findings have been evaluated.
The reading window may show a light pink background coloration; this will not affect the accuracy of the results.
The manufacturer and its distributors cannot be held responsible for the consequences of misuse or misinterpretation of the results given by the test.

Expected Values

The Rapid C.Brucella Ab Test Kit has been compared with 2-mercapto- ethanol Rapid Slide Agglutination Test. The overall accuracy is greater or equal to 90.0%.

Background

Introduction

The kit is based on sandwich immunochromatographic assay, to detect bovine brucellosis antibody in serum / plasma / blood, which is rapid, accurate and easy-to-operate.

Brucella canis is sexually transmitted by the mating of infected males and females. Brucella canis in the
female dog will live in the vaginal and uterine tissue and secretions for years, usually for life. In males, the Brucella bacteria live in the testicles and seminal fluids.
An infected male is just as contagious as the female since he can spread the Brucella bacteria via urine or semen. Transmission between dogs occurs via mucous membranes, so the bacteria may enter the body through the nose, mouth, conjunctiva of the eye and vagina.
The majority of bacteria in infected dogs are secreted in semen and vaginal secretions. Bacteria may also be present in milk, urine and saliva, thus any body fluids can
infect another dog.
When an infected bitch aborts, spread throughout a kennel can be very rapid. Infected bitches may deliver both living and dead puppies. These surviving puppies
are infected and will shed bacteria in their secretions.

WHAT IS THE IMMUNOCOMB ASSAY?

The ImmunoComb test is a modified ELISA, which can be described as an enzyme labeled “dot assay”, that detects antibody levels in serum, plasma or whole blood. The kit contains all the necessary reagents for developing the test. Results for the Brucella canis tests are obtained within 23 minutes.

HOW DOES THE IMMUNOCOMB WORK?

■ The ImmunoComb Kit contains 2 main components: a comb shaped plastic card, hereafter
referred to as the Comb and a multi compartment developing plate.
■ The Comb has 12 teeth – sufficient for 12 tests. Each tooth will be developed in a corresponding column of wells in the developing plate. Individual or multiple tests are processed by breaking off the
desired number of teeth from the Comb.
■ A test spot of Brucella antigen is attached to the lowest spot of each tooth on the Comb. The upper
most spot is a Positive Reference.
■ The first step of the test is to deposit a serum, plasma or whole blood specimen into a well in row
A of the multi-compartment developing plate.
■ Next, the Comb is inserted into the well(s) with the sample(s) and transferred to the remaining wells
(B-F) at timed intervals, according to the step by step instructions (see section VII). Specific IgG antibodies from the specimen, if present, bind to the antigen
at the test spots and will be labeled in row C, which contains an enzyme labeled anti-dog IgG antibody.
■ At the end of the developing process, a purplegrey color results are developed in all Positive Reference spots and in any positive sample tested spot.
■ The intensity of the color result corresponds directly to the antibody level in the test specimen. Results are scored using the Positive Reference spot and CombScale.
