<p>Why is it so hard to lose weight? A lot of people may have been sharing this sentiment and asking this question for years. What they don’t know is that three factors contribute to your body shape and size, and therefore make weight loss difficult.</p>

<p>The first factor is age. Naturally, people get round out a little as they get older. This is because of decreased metabolism and less movement. Also, menopause can cause women to suddenly gain weight due to alterations in hormone levels. Men and women may even find that they can no longer eat as much as they did a few years back without gaining some pounds.</p>
<p>The second factor is the set point of your weight and your metabolism. The set point of your weight is predetermined. This means that your body stubbornly clings to a certain amount of weight. Also, some people have faster or slower metabolic rates than others. This makes losing weight either easier or more difficult. For people with faster metabolism, the burning of calories is more abrupt, thus making weight loss easier to achieve.</p>
<p>The third factor is genetics. Your body shape, weight, and size are affected by your genes. They may be passed on to you from your parents, or even your parents’ parents. Sometimes, no matter how much you eat, if you are a family of big structured people, then you will find that losing weight is a greater challenge for you.</p>
<p>However, despite these three factors, it may still be the poor choices that we make in our daily lives that lead us to packing on the extra weight. We eat too much, we drink, we smoke, we live sedentary lives, and we are unable to break these bad habits even if we wanted to. Although originally mainly physiological data on bile acid synthesis  and transport were emphasized, and later on also the therapeutic  benefit of bile acids in gallstone disease and cholestasis was  discovered, we have come now to the molecular biology and genetic era  with major discoveries in transport defects and related diseases. This  book is the proceedings of Falk Symposium No. 120, held in The Hague,  The Netherlands, on October 12--13, 2000 -- the 16th  International Bile Acid Meeting.   One of the main discoveries recently has been the identification of  nuclear receptors for bile acids, which gives them a much broader  perspective than previously anticipated. It even suggests that bile  acids can regulate their biosynthesis and enterohepatic circulation  transcriptionally. It will therefore not be surprising that this  topic, together with the molecular regulation of cholesterol  7a-hydroxylase and cholesterol homeostasis, has a dominant place  in these proceedings. Another important topic is the progress in our  molecular understanding of hepatic (both at the basolateral and  canalicular sites), cholangiocytic and intestinal bile acid transport  processes. Further insights into genetic defects causing cholestasis  or intestinal malabsorption in animal models and in human diseases are  also discussed by a number of well-known authors. Finally the last  section deals with new findings on the role of bile acid therapy in  cholestatic syndromes or chemoprevention and with the potential  benefit of bile acid inhibitors. All contributors provide an update on  the most recent developments in their field.
