Synlogic backs onto Nasdaq by merging with Mirna

Synlogic has landed a Nasdaq listing and an $82 million cash pile by simultaneously pulling off a reverse merger with Mirna Therapeutics and a series C round. The activity tees up the Jose Carlos Gutierrez-Ramos, Ph.D.-helmed synthetic biology startup to push its lead candidate into the clinic in the coming months.

Cambridge, Massachusetts-based Synlogic turned to a mix of new and existing investors including Arctic Aurora LifeScience, Atlas Venture, CLI Ventures, New Enterprise Associates (NEA) and OrbiMed for the $42 million series C. That round comes 15 months after Synlogic pulled in $40 million in a series B. And, when combined with the cash Mirna has left from its failed tilt at drug development, gives the company $82 million to fuel its move into the clinic.

G1 Therapeutics thinks it has a winner on its hands with CDK4/6 inhibitor trilaciclib and is embarking on an initial public offering to give it the resources it needs to bring it to market.

The Research Triangle Park biotech says it is hoping to raise around $115 million from the IPO, which comes a little over a year after G1 raised $47 million in a venture round on the strength of some early-stage data for its lead drug.

Going public signals G1's increasing confidence in trilaciclib (G1T28), which is in the same class as Pfizer's Ibrance (palbociclib)—the first drug of this type to reach the market in 2015 and already a $2 billion-a-year blockbuster—and Novartis' new entrant Kisqali (ribociclib). Eli Lilly is also nipping at the CDK4/6leaders' heels with its abemaciclib prospect, due to report late-stage data shortly.

The FDA has granted priority review status to Bayer’s copanlisib, setting the PI3K inhibitor up to speed through the regulatory process in 6 months or less. Bayer secured the privileged position on the strength of data from a midphase trial in patients with non-Hodgkin lymphoma (NHL).

Landing the regulatory status means Bayer will learn whether the FDA shares its enthusiasm for copanlisib within 6 months, rather than the 10 months the agency typically takes to review drugs. The precedent set by other cancer drugs with priority review status suggests the FDA may make a decision about copanlisib in less than 6 months if it is particularly bowled over by the data.

Editas delays IND for Allergan-partnered CRISPR program

Editas Medicine has delayed the target date for filing an IND for its lead candidate. The setback to the Allergan-partnered CRISPR program stems from delays at a third-party manufacturer working on Leber congenital amaurosis treatment LCA10.

Cambridge, Massachusetts-based Editas had planned to file an IND for LCA10 by the end of the year. Now Editas has delayed that major moment in its short history and that of the broader CRISPR field until the middle of next year. The delay stems from a misstep in the production of a material used in the manufacture of the adeno-associated viral (AAV) vectors Editas will use to deliver its gene editing payloads.

“AAV manufacturing requires several steps to happen in perfect sequence for things to all come together. And we’re using several external contractors to perform these steps. We have to produce the input material that all comes together to then create the AAV in a cell culture systems,” Editas CTO Vic Meyer told investors.

AstraZeneca sues exec Luke Miels after his defection to GlaxoSmithKline

GlaxoSmithKline's poaching of pharma industry veteran Luke Miels from AstraZeneca in January was billed as an early win success for incoming CEO Emma Walmsley, who scrambled to replace global pharma president Abbas Hussain and other departing execs.

Fast-forward to the present, and the appointment has opened a can of worms. Bloomberg reported that Miels is currently on "gardening leave" from AZ after facing a lawsuit from his current employer alleging breach of employment.

AZ wants Miels to honor the terms of the contract he signed as executive VP of its European pharma business and has taken to the courts to try to force him to do so, according to the newswire.

Intellia moves closer to clinic with CRISPR tech

Intellia Therapeutics has taken another step toward human trials of its gene editing technology after reporting new data in animal models.

The CRISPR specialist says it has been able to show for the first time that it is able to not only achieve long-term suppression of a gene using its gene-editing CRISPR/Cas9 drug, in this case the sequence coding for serum transthyretin (TTR) protein, but also demonstrate a dose-dependent reduction in the activity of the target gene in a second animal species.

New data from studies in mice show that a previously reported 97% reduction in TTR—driven by 70% gene editing efficiency working in mouse livers—lasts for up to six months from a single dose. Meanwhile, a study in rats showed a similar dose-dependent reduction in the TTR gene, and crucially evidence of comparable activity in a second species as Intellia builds the case to move to the clinic.

The biotech industry is mourning the loss of one of the sector's founding fathers after the death of Genzyme founder Henri Termeer at the age of 71.

According to the Boston Globe, Termeer died after collapsing at his home in Marblehead, Massachusetts, on Friday night.

Termeer was at the helm of Genzyme for nearly 30 years, joining the company in 1983 and rising quickly to the role of CEO before it was acquired by Sanofi in 2011 for more than $20 billion. Prof. Dr. M Amjad Hossain was born in Shukdebpur, Dinajpur, Bangladesh in 1953. He was an young freedom fighter. Its sorrow that to express he was injured in the war of Liberation in 1971 (between Bangladesh & Pakistan) when he was only 19 years old. He was shot on his Femur and was cured by Dr. R. J. Garst. #Short story of Prof. Dr. M Amjad Hossain told himself about the moment of Freedom Fight#

In 1971, I was a young man who has just passed his intermediate exams. My father wanted me to join the army. During the tensed days of March, I was at the cantonment for my ISSB exams. The hatred of the Pakistani soldiers and their commanders towards the Bengalis and Sheikh Mujib was quite clear there. The oppression that we were under the last two decades, how the Pakistanis treated us like vermin, how they exploited us financially and politically- all these created a spark in me. After the military crackdown, it went ablaze.

After the slaughter of 25^th march, my family searched for a safe haven. A large number of non-bengali Biharis lived in an around my home Chirirbandor Dinajpur, so it wasn’t safe there. The women moved to remote villages. After geting the full information for the Pakistani Army they would not allow us to live with dignity rather they will crash the Bengalis. So my father took 25 young man and cross the border for west Dinajpur. It was 3rd week of April when the exile Bangladesh Govt. has formed under the Leadership of Taj Uddin Ahmed and Syed Nazrul Islam and the guerilla war started. We reached kamarpara camp where I was trained in guerilla warfare under Prof. Abu Sayeed and Captain Anwar. I took further training in Raygonj, Panighata and Shiliguri after I was made the leader of my team. I fought in deferent places of Dinajpur District under sector 7 and carried out operations in “Boro Gram”,” New Town” and “Rudrani”.

The battle in where I was shot took place on 31^st July in a village named “Bharam” in Fulbari. We were to ambush a military convoy at dawn. So we took position from the night before. But somehow our positions were betrayed and the army got air of us. They didn’t show up in the morning. We went inside the village for food. In the mean time, with the aid of Razakars, the army had surrounded the area and caught us off guard. My fellow mate Muzaffar took the charge of covering fire while we tried to escape. Muzaffar sacrificed himself saving the whole team. I got shot in my left femur while getting back. I would have been dead if the reinforcements hadn’t arrived just in time.

Wounded, I was sent to Balurghat Hospital and then deferent Military Hospital. I was there till the historical moment of independence. I returned to Bangladesh as a free man and in 1972 I got admitted in Dhaka Medical College. I graduated from DMC in 1978 and pursued my carrier in orthopedics. As I was shot in the femur, that was somewhat an inspiration for me for taking up orthopedics as a career as well as Dr. R. J Garst, I met in early 1972 for my treatment.

I’ve also served as the health advisor of “Muktijoddha Kalyan Trust” where I have treated many of my fellow freedom fighters. The words of Dr. Ronald Joseph Garst, my mentor, will always be my motivation for the relentless service I want to render to my beloved country-“these fighters are your other family. You are one of them. Doing something for them is the next war you must fight”

Upon completion of his graduation in Medicine in the year of 1978, Prof. Dr. M Amjad Hossain followed his professional career to orthopedics as well as successfully achieved his MS Degree from National Institue of Traumatology and Orthopaedic Rehabilitation under University of Dhaka (1986). In every sphere of his life, he has devoted his time and tried to engage and serve to the Orthopaedic department of various Institutions across the nationally and Internationally.

Prof. Dr. M Amjad Hossain also took part in the Liberation War of Bangladesh in 1971 (Sector 7). At that time, he served to many disabled and injured fighters who wounded by Pakistani Army. As well as he was shot on his femur. At a point, he was treated by DR. R.J. Garst and get cured. After the announcement of liberty he came back to Bangladesh freely with cured. As it was everything well that Bangladesh won the Freedom and achieved the Liberty but still from that moment Prof. Dr. M Amjad felt he will dedicate his rest of life for the people serving them who is becoming disabled by accident or any other natural disaster.
When he was in the Bed of Hospital with a severe pain of his Femur, he thought about the days ashed, he thought about the people and thought about the Orthopaedic.

He persuade himself with lots of training on the update issues and experience exchange with the most of leading orthopedic fellows of many countries. Among them USA, Singapore, Germany, Australia and India has actively contributed a new-era of his professional excellence today.

X-ray Text of an interventional: shows new imaging system may cut X-ray exposure for liver cancer patients

Johns Hopkins researchers report that their test of an interventional X-ray guidance device approved by the U.S. Food and Drug Administration in 2013 has the potential to reduce the radiation exposure of patients undergoing intra-arterial therapy (IAT) for liver cancer.

In a report prepared for presentation Dec. 3 at the 100th annual meeting of the Radiological Society of North America in Chicago (abstract #SSM24-02), the researchers described the results of a clinical trial of the imaging system AlluraClarity, made by Philips Healthcare, on 50 patients with liver cancer. Its use reduced radiation exposure up to 80 percent, compared with exposure from a standard imaging X-ray platform used in IAT, while producing images just as clear as the standard system, says Jean-Francois Geschwind, M.D., a professor in the Russell H. Morgan Department of Radiology and Radiological Science in the Johns Hopkins University School of Medicine and its Kimmel Cancer Center.
Geschwind says if further studies continue to affirm his team's findings, the platform may be especially useful for patients who need repeat therapy; children, who are especially vulnerable to radiation; and physicians who routinely use procedures such as IAT and are exposed to radiation.

During IAT, a physician inserts a thin, flexible tube directly into a blood vessel feeding a tumor, using that pathway to deliver chemotherapy or other drugs. X-ray imaging is used during the procedure to visualize the patient's blood vessels and to guide both the catheter's placement and drug delivery.
Geschwind and his colleagues compared the radiation exposure of 25 patients with liver cancer treated with IAT using the AlluraClarity platform to the exposure of 25 additional patients with liver cancer treated with IAT using Philips' previous X-ray imaging platform, called Allura.

Lowering the radiation power on standard X-ray imaging platforms can reduce the exposure, but without special image processing, the amount of image noise increases and physicians are unable to see small structures needed for good treatment, says Ruediger Schernthaner, M.D., a postdoctoral research fellow in vascular and interventional radiology at The Johns Hopkins Hospital. "You can compare this to an image taken with your cell phone in the evening without a flash," he says.
The AlluraClarity platform uses a series of real-time image processing algorithms to achieve high quality images at a lower radiation power, Schernthaner says.

3-D mammography improves cancer detection in dense breasts

A major new study being presented at the annual meeting of the Radiological Society of North America (RSNA) has found that digital breast tomosynthesis, also known as 3-D mammography, has the potential to significantly increase the cancer detection rate in mammography screening of women with dense breasts.

Breasts are considered dense if they have a lot of fibrous or glandular tissue but not much fatty tissue. Research has shown that dense breasts are more likely to develop cancer, a problem compounded by the fact that cancer in dense breasts can be difficult to detect on mammograms.
Other imaging modalities like ultrasound and MRI are often used to help find cancers that can't be seen on mammograms, but both modalities have higher rates of false-positive findings, which are suspicious findings that turn out not to be cancer. This higher false-positive rate often results in more tests and unnecessary biopsies, making MRI and ultrasound expensive to implement in high-volume screening programs, according to study lead author Per Skaane, M.D., Ph.D., from the Department of Radiology at Oslo University Hospital in Oslo, Norway.

Dr. Skaane and colleagues have been studying tomosynthesis as a promising breast cancer screening option that addresses some of the limitations of mammography by providing 3-D views of the breast.
"Tomosynthesis could be regarded as an improvement of mammography and would be much easier than MRI or ultrasound to implement in organized screening programs," Dr. Skaane said. "So the intention of our study was to see if tomosynthesis really would significantly increase the cancer detection rate in a population-based mammography screening program."

The researchers compared cancer detection using full-field digital mammography (FFDM) versus FFDM plus digital breast tomosynthesis in 25,547 women between the ages of 50 and 69. Breast density was classified based on the American College of Radiology's Breast Imaging-Reporting and Data System (BI-RADS). The BI-RADS breast density scale runs from 1 to 4, with 1 being the least dense and 4 being the most dense.

There were 257 malignancies detected on FFDM and a combination of FFDM and tomosynthesis in the study group, including 105 in the density 2 group and 110 in density 3. Of the 257 cancers, 211, or 82 percent were detected with FFDM plus tomosynthesis, a significant improvement over the 163, or 63 percent, detected with FFDM alone.

FFDM plus tomosynthesis pinpointed 80 percent of the 132 cancer cases in women with dense breasts, compared to only 59 percent for FFDM alone.
"Our findings are extremely promising, showing an overall relative increase in the cancer detection rate of about 30 percent," Dr. Skaane said. "Stratifying the results on invasive cancers only, the relative increase in cancer detection was about 40 percent."

Tomosynthesis not only improved the cancer detection rate in women with dense breasts, it also helped increase detection for women in the "fatty breast" BI-RADS categories. The addition of tomosynthesis to FFDM improved the cancer detection rate from 68 percent to 84 percent in women with BI-RADS density 1 or 2 breasts.
"Our results show that implementation of tomosynthesis might indicate a new era in breast cancer screening," Dr. Skaane said.

US-based Medidata has completed a method development project carried out in partnership with GlaxoSmithKline (GSK) to evaluate the impact of unifying mobile health (mHealth) devices with cloud-based technologies in a clinical trial setting.

Conducted at GSK's Human Performance Lab, the joint project evaluated the capabilities of mHealth tools and how they could be used to enable a new model for clinical trial conduct that aligns site and patient needs with faster study execution and reduced costs.

The project showed mHealth technologies can comprehensively collect large volumes of objective data that is reliable, secure and ready for analysis, providing real-time, continuous insight into patients' well being.

Complete data from the project was audited and is said to be compliant with US Food and Drug Administration (FDA) regulations.

In the project, participants were given two wearable devices Vital Connect's HealthPatch MD and ActiGraph's wGT3X-BT Monitor to continuously measure vital signs, electrocardiogram (ECG) data and activity levels.

Additionally, participants used Medidata Patient Cloud, a mobile app for patient-reported outcomes, offered as part of Medidata's technology platform.

Patients carried smartphones that captured data from mHealth devices, which pulled this data into the Medidata Clinical Cloud and then mapped it to the clinical record.

During the trial, patients were asked to continue their standard daily routine and checked in with the performance lab only at the start and end of the effort.

Unlike conventional rigid laparoscopic surgery, the SPIDER flexible laparoscopic platform allows multiple instruments to be used through one incision. The result may potentially be fewer incisions and a fast recovery for the patient, while providing surgeons with enhanced capabilities.

How does the SPIDER® Surgical System Work?

Generally spider works in following way:

The surgical device is inserted through a small incision in your belly button.
Once inside, the system expands umbrella-like, and the surgeon can use a variety of instruments to perform the procedure.
Once the procedure is complete, the surgeon collapses the system in preparation for removing.
The tools and the system are removed through the original incision.

Only 3 in 10 Americans with HIV Have virus in check

Just 30% of the 1.2 million Americans with HIV had the virus under control in 2011, according to a report released yesterday by the Centers for Disease Control and Prevention. Among those who don’t have their virus in check, two-thirds have been diagnosed, but are no longer in care, according to the report. When used consistently, antiretroviral treatment can keep HIV at very low levels in the body (known as viral suppression), allowing people with HIV to live longer, healthier lives and substantially reducing the likelihood that they will transmit the virus to others. The report says doctors, nurses and health care systems should test patients for HIV as a regular part of medical care; counsel patients who do not have HIV on how to prevent it; make sure people living with HIV are prescribed and take HIV medicines, stay in care and get supportive services; and work with health departments to get and keep people in HIV medical care.

Trans Enterics (Durham, NC), maker of the highly articulating single-site Spider laparoscopic surgical system, just released a preview video of a robotized version of the device. Manipulation of the two pincers looks pretty intuitive, but the controls for the articulating light are less so. There’s the ability to change the scale the movement of the device so that during close-up work the surgeon can gain a new level of precision with a push of a button.
Though the device is still in development, the company hopes it will be a cheaper alternative to other robotic surgical systems. We’re thinking they’re thinking of the da Vinci. Check out the SurgiBot:
