I just got my first kit in the post so here is a short overview of what you may want to know as there has been quite some development as companies are trying to
find the right business model.

How does it normally work? You order a kit, spit in it, send it back, wait (quite a bit) for data and analysis. The current WSG industry standard is 30X
sequencing that allows sufficient whole genome sequencing but may not catch de
novo mutations. For those, you may want higher coverage like 130X Whole-exome sequencing for
known targets. Generally, all options are based on 150bp paired-end reads (no worries if you don't know what that means).

so to the options:

Nebula Genomics – discounted rates start at 300$
for 30X. They have a subscription-based business model so effective costs will
be higher depending on your choice and expertise in analyzing your DNA (30X + lifetime
support is 500$). If you want to have higher confidence they now offer also a
100X WGS for around 1000-1200+$.

Dante Labs is a very attractive alternative to Nebula without
the subscription model (but higher upfront costs). Also ships internationally, additionally
offers a combined 30X WGS and 130X WES, and is a well-established leader in the
field.

Both Dante and Nebula also offer DNA analysis services if you already
have had your DNA sequenced but not yet analyzed. Oh and you can download the raw data at any time (be prepared it's a lot of data).

the first player on the market however currently an uncertain option. Due to
their financial insecurity earlier they currently lack clear info on their
options. They also have an increased barrier as they require physician approval
and access to your raw files costs extra.

To-customer DNA genome screening, DNA analysis,

Innovative Neurotech Start-ups

Neuroscience driven technology holds great promise and provides a rich resource of inspiration towards an exciting future for consumers, patients, businesses, and entrepreneurs alike. Elon Musks' recent announcement of Neuralink and its enthusiastic goals has put Neuroscience again at center stage for translating innovative research into business ideas. Here, I will provide a small glimpse into rising startups and innovative ideas that you may never have heard of.

- Note I will focus on the innovative aspect of the idea or product and not evaluate the business model, science, team, product-, market-, or financing strategy,....

Secret Chord Laboratories - makes use of the insight into how our brains appreciate music. They aim to provide an Ai-driven model to predict how well sounds, songs, and music are received by the audience. A very timely translational effort that holds great business promises and will raise the ears in the music industry and in the world of advertising. Done properly by combining the right behavioral and physiological read-out measures and optimizing deep-learning algorithm - a strategy that almost certainly will succeed. Let's just hope it will not result in an even more optimized and thus generic popular music industry but allows flexibility and noise to push forward new concepts of how we listen to music.

Spare-change Investment is the newest hype for the Populus. Currently, you can't go a day without being bombarded by acorns. However, what we should be really talking about is InvestAcure. A model that could be a way forward to speed-up translational research efforts. Similarly big aims for brain-computer interfacing, well funded and holding great potential. In stark contrast to Neuralink, Kernel focuses on non-invasive brain recording technologies in combination with state-of-the-art machine-learning and deep-learning algorithms. The heart of this recording technique relies on a novel optically pumped magnetometer sensors for measuring the magnetoencephalogram of the brain. We hope that both efforts will be successful and advance human knowledge of the brain and greatly improve the understanding and translation of neuronal signals.

"Proud of being humble" - A personal note on the legacy of Oleh Hornykiewicz

" I am only proud of being humble ", Oleh Hornykiewicz (1926-2020)

Personally,
I have to deeply thank Oleh, not only for his memories and the time he was willing
to spend with a young scientist like me, but also for a timely reminder restoring
faith in what makes a great scientist. A reminder that a successful scientist is
not measured by the prizes he or she acquires, nor for the low-hanging fruits they
collect because they can. A scientist is measured by those moments of profound breakthrough
discoveries that ripple and reshape the landscape of science. Those are the moments
of which you can be really proud to have achieved in your lifetime. And this is
the story of one of those moments....

Back in 2012, having recently received my PhD from Trinity College in Dublin, I
took up a Post-Doc in Vienna. Within the halls of the Center for Brain Research,
where Karl Landsteiner discovered human blood groups, I encountered a late octogenarian
whom I believed to be from a similar time period as Landsteiner. Intrigued by
his almost daily presence at the center, I asked my new colleagues of who that gentleman
may be and what the heck was he doing here? After more enquiries I finally
was able to spell his name correctly to allow a google search. It turned out to
be Oleh Hornykiewicz, a scientist who revolutionized Parkinson's Disease (PD) treatment
and who had such a tremendous and lasting positive impact on so many patients suffering
from this debilitating disease. A scientist with whom I had the pleasure of
many conversations throughout my years in Vienna. A scientist of old who shall not
be remembered as someone who "should have received the noble prize" but
for the outstanding scientist and person, he was.

(left) Oleh Hornykiewicz and (right) Walther Birkmayer
who solved a unique mystery underlying Parkinsons Disease

Hornykiewicz
and fellow colleagues like Walther Birkmayer described for the first time the absence
of the neurotransmitter, dopamine, specifically in the substantia nigra of patients
afflicted by PD, thereby laying a new foundation for PD research and treatment
to this day. However, as so often happens, breakthroughs in science take their time
and are often met with skepticism from fellow peers. So Oleh had to go one step
further. Being in Vienna in the early 1960’s with health regulations less stringent
than today, an idea started to form in Oleh's brain when he glimpsed a long-forgotten
bottle of L-Dopa (a precursor to Dopamine) on a shelf within the Department of
Pharmacology. L-Dopa in hand, he walked across the street to his colleague and clinician
Walther Birkmayer convincing him to give it a try on his unfortunate patients
with PD. One may have to remember that patients of those days were in terrible shape
– unable to move or control their movements, among their many other debilitating
symptoms. A few days later they were ready for the first injection of L-Dopa ever
to take place, simultaneously anxious and hopeful to observe the outcome.

What happened
next I will remember solely due to the delight and joy on the face of a 90-year
old Oleh Hornykiewicz recalling a glimpse of history. A memory for which he was
immensely grateful. Oleh had a tremendous memory and remembered my name after the
first time he heard it, later telling me that it reminded him of a long-foreclosed
Viennese Cafe and Wine House for which he had fond memories. Coincidently the namesake
Cafe that saved my grandfather from certain death in a Russian POW camp. My grandfather
could convincingly assure the guard, who had similar fond memories of the place,
that he is closely related to the owners - he wasn't - but could so gain double-rations
and better treatment that eventually should save his life.

Almost immediately following the injections it became clear that the highly experimental
treatment alleviated the symptoms of PD, it became a key moment in Oleh's life.
A moment for which every scientist strives, one that trumps all prizes. A moment
that rewards you for all the hard work and sacrifices. The magic of science
happens, when tremor stricken and bed-ridden patients regain their motor control
and are able to stand up and walk again. A joy of the moment that should stay with
Oleh forever and which he had the pleasure to encounter many times over in Austria
and later in his many successful years in Toronto. Canada should become his
home for many years, where most of his family now reside and where on the occasion
of his 90s birthday President Trudeau personally thanked him for his achievements

China: China Brain Project

From an evolutionary standpoint some psychologists believe that the optimal time for a relationship is 3-4 years as the offspring would then reach a age where the mother may not need the protection and sustenance of a partner anymore. Not surprisingly this statement is backed by hormonal changes in females during this time period of a relationship and is taken as a reason for many break-ups.

In sharp contrast to the above hypothesis, the medical field unanimously agree the optimal time to break up with a parasite is as soon as possible. A Chinese man did not head the calls and as it should turn out, has lived with a parasite for more than 4 years. Now, that is not necessarily surprising as their are a number of parasites which go unnoticed for a prolonged time period. However, in his particular case, it is surprising as the parasite was neither of small size, nor was it in a part of the body where you would expect it. In fact, the parasite worm settled in his brain - so you can imagine when doctors in Cambridge saw his brain scan images for the first time. See below what they have seen:

They finally managed to successfully remove the 1-cm long parasite. However, the patient still had long-term health effects as a result of the parasite infestation and local brain tissue damage. The parasite (Diphyllobothriidea tapeworm) mainly infects amphibious animals and fish. Consumption of raw or improperly cooked fish was probably also the reason of infestation in the presented case here.

With the Star Wars hype full on the way, many of you may not have realised that the Star Wars saga assisted us in one of the most important neuroscientific discoveries of recent years. In this particular case Star Wars not only aids us to explain one of the most central concepts of our brain but at the same time helped to discover it.

All of us learn better when we are able to connect newly acquired facts and knowledge with something we already know - a central and crucial process in learning which helps us to learn more efficiently. The same is not only true for learning but also for recalling and remembering certain things. Germans (as always) have a specific name for this: "Eselsbrücke" (donkey bridge). It goes back to the fact that the only way to get a stubborn donkey to cross a stream is to send him over a bridge. And sometimes a donkey bridge is the only way to get our stubborn memories to remember something. So essentially it is easier to remember a name of a river if you have already been there and can link the name with a memory already available in your mind.

So in order to form connections between certain elements we need to understand concepts. We would not see an obvious link, between a cow and a pencil, however between a cow and a farm or between a cow and cheese. You can also think of each concept as a bucket, once you learn something new, you decide in which buckets your new item will fit. Now what has it all to do with Star Wars. Rodrigo Quiroga and his team had a unique chance when he was allowed to record from actual neurons within the brains in awake patients. Sounds like science-fiction not really. In severe cases of epilepsy it is required to perform a brain surgery to identify the location of the seizure. This procedure requires the introduction of an electrode to stimulate certain areas. With a small tweak you can not only stimulate but also record the neuronal activity from a small and selected population of cells within the brain without any further harm or danger to the patient.

During one of those procedures (the patients are awake as they would have to respond to stimulations of the brain) they were also shown a set of pictures. After recording a number of neurons the team realised one particular neuron which increased its firing activity when the patient saw pictures of Luke Skywalker (see the figure below). Not particularly surprising, however, the same neuron responded similarly when the name Luke Skywalker was shown on screen or spoken by a female or male voice. So now you may be wondering why we do not call the neuron - The Luke Skywalker Neuron - as the neuron did not respond to any other picture of people or objects in the test set. And yet it responded to one more picture - the picture of Yoda. This proved that the neuron did not respond only to Luke Skywalker himself but characters associated with it. Due to time constraints they could not test more items and characters but in further experiments the team proved that these neurons would be able to form the basis of how we form associations and concepts and at the same time would explain why the hippocampus (a central brain structure where those cells were recorded) is so important for encoding and recalling of episodes and memories.

Those individual neurons would be able to form the building blocks of a network that would represent our bucket from earlier on. Now if Disney decides to introduce a new character (like the BB-8 droid) in the series your neurons could find anchor points in the Star Wars network/bucket. The neuronal connections to this neuronal network would strengthen everytime you recall the memory of BB-8 or experience it in another episode on or of the screen. Their finding not only awakened the Star Wars Neuron in Neuroscience but at the same time and for the first time in humans identified brain cells which could perform one of the most crucial tasks in our brains.

The response of an individual brain cell in answer to certain visual and auditory stimuli.

To quote Quiroga himself in a recent publication the characteristics of this concept cells, as he calls them as following:

These neurons have a relatively long latency, suggesting lateral processing to extract the meaning of the stimulus

They tend to fire to personally relevant concepts, namely, those that the subject may care to store in memory;

They have a high degree of invariance, which is in agreement with the fact that we tend to remember concepts and forget irrelevant details

They have a sparse, explicit and non-topographic representation, which is ideal for memory functions such as creating new associations

Their function is beyond sensory processing, given that their firing can be triggered by different stimulus modalities or internal processes in the absence of external stimulation

The new book from Prof Shane O'Mara

Turn on the TV and you will soon find a series where torture is applied not only to get confessions but information - ideally to save the life's of others (if the good guys apply it). Unfortunately this is not only seen on Television but you still find that torture techniques are not a thing of the past but are still applied all around the world. The common consensus in public or within military organisations is still that torture is effective in getting what you want. And there the problem already starts, it may not be the truth. So Prof. Shane O'Mara goes on a quest to debunk the mythical cloud around torture and explains us how extreme stressors affect the brain and lead to unreliable information.

None of those songs will probably ever get a Golden Record award, but they certainly speak to the heart of many PhD students, post docs, med studens and whoever is in research. One of those long nights alone in the lab and a new idea for a song parody was born. Here are my best 5 of them.

Nr. 1: Less known, but my favourite being a post-doc - Bohemian Rhapsody

Nr.2: For the lazy hangover day in the lab (we all had one of those)

Nr. 3: Probably the best known, at least the most viewed on youtube - Bad Project

Nr. 4 - Don't stop pipetting

Once in a while you come across those rare jewels within the medical literature. This is one of them. Here is the case of a French Casanova who made his wife his mistress. To the best of his knowledge he was cheating on his wife with a complete stranger. I guess to impress on this new women in his life he even changed his usual sexual habits (it is not reported in what ways exactly). But luckily for him his wife did not file for a divorce because she was in fact the woman who seduced him one more time.

So how could this happen? In the world of Brain research this phenomena is called Capgras Syndrome. (For the German speaking audience my previous post on the syndrome with more details and cases). People suffering from this Syndrome experience a delusional misidentification of objects, places and most of the time people. They would for example not recognise their parents in front of them but would treat them as fraudulent imposters. So Neuroscience has a good excuse if you cheat on your wife with your wife, but unfortunately I have to tell you, we still have no good excuse if you cheat on your wife with somebody else. That case won't get you into the medical literature. It will only get you straight into the hands of a divorce lawyer.

- a rare non lethal disease which we do not wish on anyone. Dissecting Scalp Cellulitis as the name implies results in an inflammation of the scalps hair follicles and an accompanying skin irritation. The causes are not entirely known, but as DSC often appears with acne conglobata and hidradenitis suppurativa it hints towards a common pathogenic mechanism of abnormal follicular keratinization and occlusion interlinked with bacterial infections. In addition, 90% of all patients are black men mostly in their 20s to 40s. In belows case however we have a white man who has suffered incredibly under the disease and thus has chosen the most extreme treatment options available. - He opted for a scalp replacement therapy as you will see in the pictures below. I WARN you before starting the video or clicking on his photobook. The pictures are very graphic and if you are not used to seeing them, they are quite disturbing.

As the disease is chronic and relapsing most of the treatment is supportive and tries to deal with pain and inflammation relief. In several cases tumor necrosis factor blockers, selective hair follicle removement and acne relief medication Isotretinoin have been utilised effectivly but conclusive data is yet insufficient as relapses might occur after many months and are not reported. In severe cases a scalp replacement is the last resort. In this case skin tissue grafted from the patient's leg has successfully been implanted onto the patients skull as you can see in the supporting pictures. We wish him the best and DSC is a thing of the past for him

Imagine you are again a 5-year old playful child. And one time your mum will take you to a medical doctor who will perform a lobotomy? Well let's hear some thoughts of such a case. If you need a short update on the method of lobotomy I suggest a quick glance at the Wiki site or check out the second documentary below.

Today I will deviate quite a bit from Neuroscience and wander into tissue engineering as I just want to share a very informative and impressive video about heart transplants and engineering. Growing organs "out of the Petri dish" and successfully implanting them into the human body has already been achieved for several organs. However, most of those organs are of relatively simple architecture. Complex organs like the heart are causing much more headaches for researchers. Hence, they had to come up with new approaches to tackle the problem. Instead of growing the organ completely new, they took a heart from a recently diseased person, decellularized the heart and then repopulate the organ with functional stem cells. But the donor heart don't need to be human, it can be also from a pig. In fact, pig organs have been successfully implanted. But as with pig hearts or hearts from donor patients the main problem up to now has been the auto-immune reaction of the patients immune system. So it is not uncommon that the patients' body often rejects the donor graft. With the new technique this problem will be overcome. However, there is still a long way to go to fully regrow a new heart but for the moment even rebuilding and re-implanting parts like a heart valves is an important step forward. Check out now the video for a better picture...!
