NIH Scientists Advance Universal Flu Vaccine

A universal influenza vaccine — so-called because it could potentially provide protection from all flu strains for decades — may become a reality because of research led by scientists from the National Institute of Allergy and Infectious Diseases (NIAID), part of the National Institutes of Health.
In experiments with mice, ferrets and monkeys, the investigators used a two-step immunization approach to elicit infection-fighting antibodies that attacked a diverse array of influenza virus strains. Current flu vaccines do not generate such broadly neutralizing antibodies, so they must be re-formulated annually to match the predominant virus strains circulating each year.
The research, led by NIAID scientist Gary J. Nabel, M.D., Ph.D., appears online ahead of print July 15 in Science Express.

"Generating broadly neutralizing antibodies to multiple strains of influenza in animals through vaccination is an important milestone in the quest for a universal influenza vaccine," says NIAID Director Anthony S. Fauci, M.D. "This significant advance lays the groundwork for the development of a vaccine to provide long-lasting protection against any strain of influenza. A durable and effective universal influenza vaccine would have enormous ramifications for the control of influenza, a disease that claims an estimated 250,000 to 500,000 lives annually, including an average of 36,000 in the United States."

In parallel experiments with mice, ferrets and monkeys, Dr. Nabel and his colleagues first primed the animals’ immune systems with a vaccine made from DNA encoding the influenza virus hemagglutinin (HA) surface protein. After being primed with DNA vaccine, the mice and ferrets received a booster dose of the 2006-2007 seasonal influenza vaccine or a vaccine made from a weakened cold virus (an adenovirus) containing HA flu protein. Monkeys were boosted with the seasonal flu vaccine only.

This prime-boost vaccine stimulated an immune response to the stem of the lollipop-shaped hemagglutinin of influenza virus. Unlike HA’s head—which mutates readily, allowing the virus to become unrecognizable to antibodies—the stem varies relatively little from strain to strain. In principle, Dr. Nabel explains, antibodies generated against the stem of HA should be able to recognize and neutralize multiple flu strains.

Although the DNA in the priming vaccine was derived from a 1999 circulating flu virus, all the animals made antibodies capable of neutralizing virus strains from several other years. Mice and ferrets produced antibodies not only against virus strains dating from before 1999, including a strain that emerged in 1934, but also against strains that emerged in 2006 and 2007.

Moreover, although the prime-boost vaccines were both made from H1 subtypes of influenza A virus, the antibodies they generated neutralized other influenza subtypes, including H5N1 (avian influenza) virus. This indicates that a prime-boost strategy potentially could confer immunity to many or all subtypes of influenza A, says Dr. Nabel.

In another set of experiments, the scientists measured how well the prime-boost vaccine protected mice and ferrets from infection with deadly levels of flu virus. Three weeks after receiving the boost, 20 mice were exposed to high levels of 1934 flu virus, and 80 percent survived. Mice receiving DNA only, seasonal flu vaccine only or a sham prime-boost vaccine all died.

The researchers saw similar results when they tested several prime-boost combinations in ferrets, which are considered a good animal model for predicting flu vaccine efficacy in humans. All four ferrets that received a DNA prime-seasonal boost were protected from infection with a 2007 virus strain, while all six ferrets that received the DNA prime-cold virus boost combination were protected from the 1934 influenza virus.

Collaborators on these studies included Terrence Tumpey, Ph.D., of the Centers for Disease Control and Prevention.

“We are excited by these results,” says Dr. Nabel. “The prime-boost approach opens a new door to vaccinations for influenza that would be similar to vaccination against such diseases as hepatitis, where we vaccinate early in life and then boost immunity through occasional, additional inoculations in adulthood.”

Trials of prime-boost influenza vaccines assessing safety and ability of the vaccine to generate immune responses are already under way in humans, Dr. Nabel adds. The information from the new research will be valuable in selecting candidates to move forward into large-scale trials, he says. “We may be able to begin efficacy trials of a broadly protective flu vaccine in three to five years.”

Hayward, a geologist who has spent his entire career at BP, recognizes that he has become "a liability going forward" and is ready to step down, the source close to the company said. The source asked for anonymity because the company has not yet announced its intentions.
Hayward has come under sharp criticism from members of Congress, President Obama and many investment analysts for events leading up to the exploration well blowout. Some of his comments about the spill have been called insensitive to gulf coast residents.

Moreover, the company has struggled to stop not only the flow of oil but the flow of bad publicity, from complaints about the speed of payment of claims to gulf residents to doctoring photographs on its Web site. Some analysts say Hayward's exit would be a statement that someone is taking full responsibility.
One of the front-runners to succeed Hayward would be Robert Dudley, a Mississippi native who joined BP from Amoco after the two companies merged in 1998. Dudley, who was just put in charge of leading gulf coast cleanup efforts, would be the first American to run the company originally known as British Petroleum.
Another source who spoke on condition of anonymity said that other top executives might also be ousted, but the source close to the company said that decision would be left to the new chief executive.
Among the executives whose positions are widely considered to be tenuous are Andy Inglis, the Houston-based head of global exploration and production who is also leading the relief well effort; Doug Suttles, the chief operating officer who has played a prominent public role in addressing questions about BP's response to claims and cleanup issues; Lamar McKay, the president of BP America who had no responsibility for the exploration well but who has been given the unenviable task of defending the company before Congress.

A serial sex offender was today sentenced to life in prison after he admitted kidnapping, raping and murdering a teenager whom he ensnared using a fake profile on Facebook.
Peter Chapman was told he must serve at least 35 years in prison by judge Peter Fox today, following the killing of Ashleigh Hall, who was 17.

The judge said he was "a very great danger to young women, and for what it is worth, I cannot foresee your release".
Sentencing him at Teesside crown court, the judge said Chapman had been the architect of "an evil scheme, very carefully brought, and with considerable detail, to trap your victim".
The killer had claimed to be a handsome teenager and used the profile to lure the teenager into meeting him after he befriended her through Facebook.
In reality, he was an emaciated, 33-year-old convicted double rapist, who was almost toothless and living in his car.
Graham Reeds QC, for the prosecution, told the court Chapman had claimed to be called DJ Pete or Pete Cartwright.
"The photograph is not of him. It is of a barechested and good-looking boy, who is apparently in his late teens."
Chapman was, in fact, "a somewhat plainer-looking man who could pass for being rather older than his 33 years. When she met him on 25 October last year, he kidnapped, raped and murdered her."
The prosecutor described how the teenager suffered from low-esteem.
The night before Ashleigh's body was found, she had told her mother she was going to stay with a friend.
In order to entice her into his car, Chapman sent her a text message saying the boy's father would pick her up. He texted her to say: "My Dad's on his way, babe."
She replied: "He's here, babe." Once in the car, he attacked her.
Ashleigh's body was found dumped in a farmer's field near Sedgefield, County Durham, in October. Her arms were bound and tape was put over her face, suffocating her to death.
Chapman was arrested by chance in a separate police inquiry after a nationwide alert went out to trace his car after he had failed to comply with the requirements of his sex offenders' registration. During initial interviews he told police they could "crush his car". He later lost his nerve and confessed to murder.
Seen on CCTV at Middlesbrough police station, he tells a custody officer: "I killed someone last night. I need to tell somebody from CID where the body is. It hasn't been reported yet."
Detective Inspector Mick Callan, head of Durham police's major crime team, said: "The truth is he is a predatory sex offender who, through the tentacles of the internet, could reach out to the young and the vulnerable.
"He knows full well that using his real name and picture would not grab the attention of any young woman for a moment. But by using the picture of a good-looking young man as a cover he has woven a web of attraction and deceit that has sadly had a murderous ending."
Chapman, who was brought up by his grandparents in Stockton-on-Tees, has a history of sexual offending. He was the subject of several sexual assault investigations, beginning when he was 15. Four years later, in 1996, he received a seven-year prison sentence for raping two prostitutes at knifepoint during separate incidents and was released in 2001.
The following year Cheshire police questioned him over the rape and kidnap of a prostitute in Ellesmere Port. The case was discontinued.
Speaking outside court Andrea Hall, Ashleigh's mother, said the authorities should have done more to monitor Chapman's whereabouts after his release from prison. "They are the sort of people who should be tagged and they should be kept an eye on all the time. They shouldn't be allowed out into society anyway."
Mrs Hall, 39, also said it was wrong that Chapman, as a registered sex offender, was able to post online as a teenage boy.

"He took somebody else's photo and put it on the internet and has been posing behind this photo. It is awful to think that there's actually a boy out there and he is using his photo to prey on young girls."

Mrs Hall, who has three other daughters, said she was powerless to stop Ashleigh going out on the night she met Chapman. "What could I have done?" she asked. "She was 17. You don't stop your kids from going out."
She recalled the horror of hearing a police officer answer her daughter's phone, having attempted to contact her mobile on 30 occasions. She said: "I could understand it if Ashleigh had died because of illness but to actually have someone take someone's life is just unbearable."

A police officer in Hot Springs, Arkansas, was photographed actually choking a thirteen-year old boy before arresting him, and several others (which was captured on video), seemingly for... skateboarding.

A video is circulating the likes of MySpace and YouTube wherein several young boys- and one girl- in Hot Springs, Arkansas, decided to celebrate Go Skateboarding Day. While skateboarding downtown, one was arrested- and it appears to be because he was skateboarding.

One youth took a picture of the Officer Joey Williams on top of thirteen year old Jarad Graham, who was facing up, and Williams has his hand around the boy's throat. After handcuffing the child, he chased the person who took the photograph, leaving the child in handcuffs. Williams later comes back, takes the child to speak with him away from his friends, and tells the boy he was in violation of city ordinance and resisting arrest... resisting arrest for what? WHY was he arrested in the first place?

When his friends come to listen, Officer Williams tells them all to go away, and that they're all in violation of city ordinance as well. He then proceeds to arrest several more people, taking the video camera and setting it on the sidewalk.
