Het CRL wordt geleid door Prof. Jan Tavernier en bestaat uit een 30-tal medewerkers (www.crl-mappit.be). &lt;br /&gt;Een belangrijk onderzoeksdomein van het CRL is het ontwikkelen van nieuwe technologieën (o.a. MAPPIT en KISS) voor de identificatie en analyse van eiwit-eiwit en chemische molecule-eiwit interacties. Voor de ondersteuning van een aantal projecten in deze context is het CRL op zoek naar een enthousiaste laboratoriumtechnoloog. &lt;br /&gt;Onder de supervisie van een postdoctoraal onderzoeker en in een team van ervaren laboratoriumtechnologen zal de geselecteerde kandidaat betrokken worden bij uiteenlopende onderdelen van deze projecten. Technieken die daarbij zullen toegepast worden zijn o.a. recombinant DNA technologie (PCR, plasmied constructie, DNA zuivering,…), eiwit technieken (Western blot,…) en celcultuurtechnieken (transfecties, FACS analyse, luciferase rapporteertesten,…).&lt;/p&gt;

To this extend, state-of-the-art high throughput technologies are being used, including large-scale next-generation sequencing of DNA and RNA samples. One of the most exciting ongoing projects concerns the analysis of whole-genome sequencing data of tumor samples. Additionally, a number of projects involve the analysis of ChiPSeq data to characterize epigenetic modifications of (tumor) genomes and based on these data identify novel transcriptional regulatory networks. A key aspect in each of these projects is the use of specialized bio-informatics for primary data analyses (sequence assembly, mapping of deletions, amplifications, tandem duplications, invertions and interchromosomal rearrangements, etc), the development of novel algorithms for specific research questions, data interpretation (prioritization of identified genetic variants through data fusion, advanced pathway analyses and the application of advanced modeling approaches to predict disease-relevant characteristics). Additionally, each of these projects also involves the functional validation of the genetic/bio-informatic findings in models organisms using standard molecular biology and functional genomic tools.&lt;/p&gt;

As cell division is pivotal for plant growth, understanding how cell cycle progression is controlled may offer ways to increase the yield of important crop species. Unidirectional and irreversible progression through the cell cycle is mediated through the selective proteolytic control of both positive and negative regulators of the cell cycle. A major protein complex dedicated to the basic cell cycle control is the Anaphase-Promoting Complex/Cyclosome (APC/C), targeting proteins for destruction. Activity of the APC/C links to the timing of cell differentiation and growth. Recently, we have identified key regulators of the APC/C (Heyman et al., 2011; Eloy et al., 2011), however how this complex controls growth and development remains elusive. The proposed project aims to fill this knowledge gap through the identification of growth related APC/C targets. To meet this objective, two independent strategies will be followed. A first strategy involves the in planta identification of suppressor genes that abolish the growth-inhibiting effect of an APC/C mutant that shows a retarded growth. The second strategy relies on a differential proteomic screen. Potential APC/C targets will be analyzed for their growth promoting effects, first in Arabidopsis thaliana, and subsequently in maize.&lt;br /&gt;&lt;/p&gt;

Our multidisciplinary team of about 70 researchers is housed in new laboratory facilities and combines extensive expertise in structural biology (X-ray crystallography, NMR, EM and atomic force microscopy), biophysics, enzymology and molecular biology.&lt;br /&gt;&lt;br /&gt;Project description: &lt;br /&gt;The research topic deals with the structural and functional characterization of newly identified enzymes involved in the acquisition of the persistence phenotype in the human pathogen P. aeruginosa. Persisters are highly multidrug-tolerant cells that constitute a small fraction of a population and are largely responsible for biofilm tolerance to antibiotics. Through collaboration with the group of Prof. J. Michiels (Centre of Microbial and Plant Genetics, KULeuven), we will combine the genetic analysis of persistence, with detailed biochemical, biophysical and structural in vitro studies, in order to contribute to a complete mechanistic understanding of the principles underlying persistence. &lt;br /&gt;&lt;br /&gt;During your PhD you will be studying the functioning and regulation of these enzymes and their interaction partners, using an approach of X-ray crystallography, enzyme kinetics and different biophysical methods. &lt;br /&gt;&lt;br /&gt;Start date&lt;br /&gt;The position is available immediately.  &lt;/div&gt;

It is a joint basic research initiative, set up by VIB, imec and KU Leuven to understand the function of brain circuits. At NERF basic neuroscientific questions are addressed through collaborative, interdisciplinary research, combining nanoelectronics with neurobiology. The center intends to push the boundaries of science, by zooming in on the functioning of neural circuits at an unprecedented level of detail. In the long run, NERF will generate new insights into the functional capabilities of the circuits of the mind as well as into research methodologies and technologies for medical applications, i.e. diagnostics and treatment of disorders of the central and peripheral nervous system (see www.NERF.be).&lt;br /&gt;&lt;br /&gt;NERF is housed on the imec campus in Leuven, Belgium, where researchers work in cross-disciplinary teams, benefitting from imec’s state-of-the-art clean room infrastructure and a new 800 m² well equipped neuro-science lab, dedicated to NERF. NERF has already recruited 3 teams of top-notch researchers doing worldclass basic research in neural circuit analysis and is recruiting 2 additional groups. Different γ-Secretase complexes exist that can regulate specific cognitive functions (Dejaegere al., PNAS, 2008; Serneels et al., Science, 2009). In addition to the production of Amyloid-beta, γ-Secretase controls other proteins that are important for neuronal function. Among them, we found APP, Notch and the schizophrenia risk genes Nrg1 and ErbB4 (Fazzari et al., Nature, 2010). However, the physiological role of γ-Secretase in the brain is still poorly understood. In this research project, we aim to determine the specific role of diverse γ-Secretase complexes in the maintenance of brain circuits.&lt;br /&gt;&lt;br /&gt;The job will involve electrophysiological recordings in different mutant γ-secretase mice available in the lab in combination with selective genetic experiments at the cellular level using viral transduction to inactivate specific gamma-secretase complexes in different areas of the brain. There is ample opportunity to interact with a multidisciplinary and international team of dedicated scientists to unravel the molecular and cell biology role of the different γ-secretases.&lt;br /&gt;&lt;br /&gt;We offer an excellent postdoctoral position in an international research institute in the center of Europe (close to Brussels and fast train connections to Paris, London, Cologne and Amsterdam), with access to high end technology, core facilities, technical support and a brand new research building. We have recently embarked on a long-term project to identify critical intracellular signaling pathways that control this process, using biochemical analysis of acutely isolated astrocytes. Ultimately, our goal is to assess the importance of identified pathways in vivo, through the production and analysis of novel transgenic mouse lines.&lt;br /&gt;&lt;br /&gt;The Project:&lt;br /&gt;Primarily, the successful candidate will be tasked with developing novel genetic tools that facilitate transgenic animal production. In particular, we are interested in the use of in utero electroporation and viral delivery techniques. &lt;br /&gt;&lt;br /&gt;In addition, a successful applicant would have the possibility to participate in the subsequent analysis of the resulting mice. Neurotransmitters are initially stored in small membranous organelles called synaptic vesicles. These vesicles are trafficked to the presynaptic plasma membrane where they fuse and release their contents.&lt;br /&gt;&lt;br /&gt;To ensure precise control of the process, the presynaptic terminal possesses a complex network of interacting proteins. The protein Bruchpilot has been identified as central to the release process in Drosophila melanogaster. Bruchpilot is thought to be the main protein component of the so-called ‘T-bar’. The T-bar is an electron dense structure found at the release site. It is thought to operate by capturing vesicles from the cytoplasm and channelling them to release sites on the plasma membrane. Importantly, we have recently identified several mutations in a region of Bruchpilot that affect the binding of synaptic vesicles to this protein. &lt;br /&gt;&lt;br /&gt;The Project:&lt;br /&gt;In this project, we will use advanced biochemical techniques to investigate the precise nature of Bruchpilot-synaptic vesicle binding. These approaches will center predominantly on novel techniques that have been developed by the host laboratory for the fractionation and analysis of Drosophila neural tissue. &lt;br /&gt;&lt;br /&gt;Following the identification of putative vesicle binding domains, the findings will then be validated in vivo using Drosophila mutants in combination with super-resolution imaging (STED and electron microscopy) and electrophysiology. &lt;br /&gt;&lt;/div&gt;

On the other hand, inflammatory caspases may dampen immune responses. &lt;br /&gt;&lt;br /&gt;To better understand the functions of inflammatory caspases, we are currently looking for a motivated Ph.D. student to work on the identification and functional characterization of inflammatory caspase substrates. The successful candidate will make use of positional proteomics technologies to determine and compare the optimal protein substrate recognition sequences of the inflammatory caspases-1, -11 and -12. We are offering a project aiming at the biochemical dissection of signaling pathways in astrocytes. &lt;br /&gt;&lt;br /&gt;Background:&lt;br /&gt;Astrocytes are the most abundant cell type in the mammalian brain, where they are known to play an essential role in development, maintenance and function of the nervous system. However, the precise molecular pathways driving these processes remain to be elucidated.&lt;br /&gt;&lt;br /&gt;The Project:&lt;br /&gt;In this project, we plan to analyze the protein and lipid signaling pathways involved in astrocyte function – by applying powerful mass-spectrometry based methods. The function of identified proteins will then be investigated using in vitro recombinant protein methods and tissue culture techniques.&lt;/div&gt;
