The first study of DNA methylation causing alternative splicing in cancer cells

Inhibiting the growth and accelerating the death rate of breast cancer cells may be possible by starving the cancer cells of glucose or by inhibiting the energy production process (aerobic glycolysis). Aerobic glycolysis confers a proliferative advantage to the cancer cells. The results were published in the Proceedings of the National Academy of Sciences.

A team of researchers led by Dr. Sanjeev Shukla from the Department of Biological Sciences, Indian Institute of Science Education and Research (IISER), Bhopal, has been able to reverse aerobic glycolysis by inhibiting DNA methylation or reducing the expression of BORIS (Brother of Regulator of Imprinted Sites) gene.

Alternative splicing

In normal cells, exon 9 gets included while exon 10 is excluded when precursor messenger RNA (pre-mRNA) is spliced or edited into mature messenger RNA (mRNA). The researchers found that splicing was different in the case of cancer cells — instead of exon 9, exon 10 gets included when the pre-mRNA is spliced into mRNA. “The methylation at exon 10 allows BORIS to bind to the exon 10 and leads to the inclusion of exon 10 in the mature mRNA,” says Dr. Shukla. “Due to aberrant alternative splicing, exon 10 gets included in the mature mRNA leading to the formation of PKM2, a cancer-specific isoform of pyruvate kinase.”

The PKM2 isoform is seen in cancer cells and is responsible for proliferation of cancer due to aerobic glycolysis while the PKM1 isoform is found in normal cells. The DNA methylation along with BORIS regulates the switch from PKM1 to PKM2 isoform.

This is the first time DNA methylation causing alternative splicing in cancer cells has been studied. DNA methylation also silences tumour suppressor genes.

“The BORIS gene is predominantly expressed in germ cells and gets over-expressed in cancer cells but not in somatic cells. So BORIS is a potential target for cancer therapy,” says Dr. Shukla.

The team studied two breast cancer cell lines and found increased exon 10 in both cancer cell lines but exon 9 was found to be increased in normal, primary cells. The presence of exon 10 in breast cancer cell lines led to higher expression of cancer-specific PKM2 isoform.

Unlike in normal breast tissue, the researchers found overexpression of BORIS in breast cancer subtypes in the Cancer Genome Atlas and Oncomine.

Potential targets

Inhibition of DNA methylation led to reduced binding of BORIS at exon 10 leading to reduced exon 10 and increased exon 9 in the mature mRNA. There was also a reduction in the cancer-specific PKM2 isoform and increase in the PKM1 isoform.

Similarly, depletion of BORIS gene led to reduced BORIS binding and decreased inclusion of exon 10 with a concomitant increase in exon 9 inclusion.

To rule out other factors responsible for alternative splicing of PKM, the researchers mutated the BORIS binding site at the PKM exon 10 region. “There was reduced exon 10 and increased exon 9 in the mutant cells. Also, there was reduced BORIS enrichment which resulted in reduced exon 10 inclusion in the mutant cells,” Dr. Shukla says.

As predicted, BORIS depletion reduced the consumption of glucose in breast cancer cells. Inhibition of DNA methylation, too, decreased glucose uptake. Interestingly, BORIS depletion went beyond reduction in glucose consumption — it resulted in reduced cancer cell growth and cell viability.

“When BORIS is down regulated it affects alternative splicing of several genes which are associated with cancer hallmark (cancer growth, reduced apoptosis abnormal metabolism). So targeting BORIS will have a great impact for cancer therapy,” says Smriti Singh from the Department of Biological Sciences, IISER, Bhopal, and the first author of the paper. “BORIS binds after DNA methylation. So we must determine whether inhibiting DNA methylation or BORIS is more effective in cancer therapy.”

Herpes treatment could be the key to curing deadliest brain cancers

THE solution to stopping an incurable brain tumour could lie with a drug now used to treat a common herpes virus.

Australian scientists are hoping to trial this, but desperately need funding from the government.

Cytomegalovirus (CMV) is a member of the herpes family that is present in half of all adults, although it shows no signs or symptoms as a healthy immune system keeps it in check.

In a major breakthrough, researchers at Duke University in the US have found this virus concentrates in the lethal brain tumour glioblastoma. By using a vaccine to attack the CMV, the Duke University trial has prolonged the lives of the trial subjects four-fold.

Three of the 11 patients are still alive and others lived for more than five years, compared to the average of 15 months post-diagnosis with glioblastoma.

University of NSW Associate Professor Kerrie McDonald said CMV proteins were abundant in glioblastoma tumours but absent in surrounding brain cells.

Her team has found a peptide (an amino acid) that can attack the CMV proteins and therefore attack the tumour without harming surrounding brain tissue.

“Because CMV concentrates in the tumour, it is a viable target and it’s a small peptide so it crosses the blood-brain barrier,” Prof McDonald said.

Scientists reveal the relationship between sugar, cancer

A nine-year joint research project conducted by VIB, KU Leuven and VUB has led to a crucial breakthrough in cancer research. Scientists have clarified how the Warburg effect, a phenomenon in which cancer cells rapidly break down sugars, stimulates tumor growth. This discovery provides evidence for a positive correlation between sugar and cancer, which may have far-reaching impacts on tailor-made diets for cancer patients. The research has been published in the leading academic journal Nature Communications.

This project was started in 2008 under the leadership of Johan Thevelein (VIB-KU Leuven), Wim Versées (VIB-VUB) and Veerle Janssens (KU Leuven). Its main focus was the Warburg effect, or the observation that tumors convert significantly higher amounts of sugar into lactate compared to healthy tissues. As one of the most prominent features of cancer cells, this phenomenon has been extensively studied and even used to detect brain tumors, among other applications. But thus far, it has been unclear whether the effect is merely a symptom of cancer, or a cause.

Sugar awakens cancer cells

While earlier research into cancer cell metabolism focused on mapping out metabolic peculiarities, this study clarifies the link between metabolic deviation and oncogenic potency in cancerous cells.

Prof. Johan Thevelein (VIB-KU Leuven): "Our research reveals how the hyperactive sugar consumption of cancerous cells leads to a vicious cycle of continued stimulation of cancer development and growth. Thus, it is able to explain the correlation between the strength of the Warburg effect and tumor aggressiveness. This link between sugar and cancer has sweeping consequences. Our results provide a foundation for future research in this domain, which can now be performed with a much more precise and relevant focus."

Yeast as an advantageous model organism

Yeast cell research was essential to the discovery, as these cells contain the same 'Ras' proteins commonly found in tumor cells, which can cause cancer in mutated form. Using yeast as a model organism, the research team examined the connection between Ras activity and the highly active sugar metabolism in yeast.

Prof. Johan Thevelein (VIB-KU Leuven): "We observed in yeast that sugar degradation is linked via the intermediate fructose 1,6-biophosphate to the activation of Ras proteins, which stimulate the multiplication of both yeast and cancer cells. It is striking that this mechanism has been conserved throughout the long evolution of yeast cell to human.

"The main advantage of using yeast was that our research was not affected by the additional regulatory mechanisms of mammalian cells, which conceal crucial underlying processes. We were thus able to target this process in yeast cells and confirm its presence in mammalian cells. However, the findings are not sufficient to identify the primary cause of the Warburg effect. Further research is needed to find out whether this primary cause is also conserved in yeast cells."

Immunotherapy is the newest weapon in the fight against cancer

The human immune system is both clever and powerful. But it is often foiled by cancer’s wily ways.

A new approach to cancer treatment — immunotherapy — aims to unmask the disease for the deadly threat it is, then direct the full force of the immune system on malignancies that would otherwise grow and spread unchecked.

Our multilayered immune defenses spot most foreign invaders and crush them decisively. But cancer, arising out of one or more mutations in our DNA, is a home-grown threat with a deceptively reassuring look. Even as cells multiply and spread, malignant cells cloak themselves in innocent garb.

Confused, the immune system pulls back its talons. And cancer has its way with us.

Immunotherapy can change this.

It stems from a growing understanding of how cancers start, grow and protect themselves from both medicines and our bodies’ natural defenses. It also capitalizes on researchers’ efforts to unpack the exquisite complexity of the immune system, including when and why it retreats from a fight.

The first generation of immunotherapy drugs is designed to release the “brakes” that inhibit the immune system from attacking cancers. By various means, they block an antibody that spares cancer cells from a sentence of “programmed death” issued by the immune system.

These so-called PD-1 blockers are the first of a class of immune checkpoint inhibitors. They are used to treat a wide range of advanced malignancies, including melanomas and cancers of the lung, breast, colon, bladder, thyroid and endometrium.

The first checkpoint inhibitor, a medication called Yervoy (ipilimumab), was approved for U.S. patients in 2011. The drug improved survival time for patients with metastatic melanoma from an average of six months to 10 months. Next came Opdivo (nivolumab) in 2014, which extended survival for patients with certain head and neck cancers, small-cell lung cancers, Hodgkin lymphoma, and some advanced colorectal cancers.

Half a dozen more immunotherapy drugs followed, including Keytruda(pembrolizumab) for lymphoma, metastatic head and neck cancers, non-small cell lung cancers and advanced melanoma.

These drugs tend to help more patients than standard therapy does, and with less punishing side effects. They extend average survival in large groups of patients — sometimes just by months, but generally by spans that are considered meaningful.

And for a small subset of patients whose prognosis is bleak, these drugs can drive cancer into outright remission. This is what instills the greatest hope in cancer doctors.

The approach doesn’t work for everybody. In rare cases, it sends the immune system into overdrive — a complication that can be uncomfortable at best and fatal at worst. This summer, the FDA halted a clinical trials in which Keytruda was being tested in conjunction with some other immune-boosting drugs to treat multiple myeloma, citing an increased risk of death.

The prices for these drugs is another drawback. Both Opdivo and Keytruda can cost $150,000 per year, a bill insurers are reluctant to pay and one that makes bioethicists concerned about unequal access.

Still, the drugs’ overall impact is clear, said Dr. Antoni Ribas, a melanoma specialist at UCLA who has conducted clinical trials on Keytruda. Immunotherapy drugs, he said, “are changing how we treat cancer.”

Just ask Kathy Thomas.

Five years ago, she was fighting a losing battle against metastatic melanoma that had spread to her breast, brain, lungs and liver. Her doctors feared she wouldn’t live to see the birth of her grandson.

Then she enrolled in one of Ribas’ clinical trials of Keytruda. Monthly infusions have made her cancer undetectable.

Last month, she celebrated her grandson’s fifth birthday.

“I would not be here if not for this drug,” said Thomas, 62.

Her experience is hardly unique. In an ongoing trial called Keystone, drug maker Merck & Co. reported this summer that close to 42% of subjects with metastatic melanoma who received Keytruda were still alive four years into the study. Some 13% of the subjects who received Keytruda had a “complete response,” and most had ceased taking the medication.

When Ribas started as an oncologist 18 years ago, “maybe one in 20 patients” lived for more than a year after being diagnosed with metastatic melanoma, he said. Now, thanks to immunotherapy drugs, “more than a third of my patients are living normal lives.”

Some cancers have proved more resistant to immunotherapy than others. Those that tend to afflict children or have weak ties to modifiable risk factors like smoking, obesity, poor diet and pollution can’t be stopped just by lifting the immune system’s brakes. To fight these cancers, the body seems to need a more specific assist.

Enter CAR-T cells.

For this approach, doctors start by harvesting a cancer patient’s T-cells, the warriors of the immune system. Scientists genetically engineer the cells to home in on the patient’s cancer and then grow millions of the modified cells in the lab. When the cells (now called chimeric antigen receptor cells, or CAR-T cells for short) are returned to the patient, they are much better equipped to hunt down and kill the cancer cells wherever they may hide.

So far, only one CAR-T immunotherapy treatment has been approved for use by the Food and Drug Administration — Kymriah, a drug designed to treat children and young adults with B-cell acute lymphoblastic leukemia who didn’t respond to standard treatment, or who suffered a relapse. Researchers tested it in 88 such patients ages 3 to 23, and it produced remission in 73 of them.

When FDA Commissioner Scott Gottlleib announced Kymriah’s approval in August, he predicted that many more CAR-T immunotherapies would follow. Dozens are currently in the pipeline, mainly aimed at treating cancers of the blood and lymph systems.

Dr. Crystal L. Mackall, a pediatric cancer specialist at Stanford, predicts that such “living drugs” will be “the beginning of a new era in medicine.” She calls them “the next frontier” in fighting cancer.

Pills and injections can target the processes by which cancers start and grow, said Mackall, who directs Stanford’s Parker Institute for Cancer Immunotherapy. But the effects of these treatments inevitably wane, and they must be renewed with another pill or injection.

“Cells can do things that drugs can’t do,” she said. “We’ve seen the dramatic effects of turning your cells into small machines, and this is just the beginning. They can be so much more sophisticated.”

Dr. Svetomir Markovic, an immunologist at the Mayo Clinic in Rochester, Minn., who specializes in melanoma, shares Mackall’s optimism.

“In many ways, we’re at the end of the beginning of immunotherapy: There’s clear benefit, but it’s still a minority of patients that get long-term benefit,” Markovic said. “We will get better at this.”

Now Momoa is opening up about the incident in a statement on Instagram.

“I awoke in Australia to the justified reactions by many people to a distasteful joke I made years ago in Hall H for which I am sorry,” Momoa said. “I am still severely disappointed in myself at the insensitivity of my remarks that day. I know my sincerest apology now won’t take away those hurtful words. Rape and sexual harassment can reach anyone and I have seen first hand its painful torment among members of my own family and friends.”

He continued: “I made a truly tasteless comment. It is unacceptable and I sincerely apologize with a heavy heart for the words I said.”

Memphis football DL Ernest Suttles' rape charge casts a pall over Tigers win vs Navy

At 2:21 Saturday afternoon, the Memphis police department tweeted the news: “Ernest Suttles was charged with rape.”

Until that moment, Suttles was a senior defensive lineman for the University of Memphis.

Less than half an hour later, Memphis kicked off a football game against Navy.

It was a massive win for the Tigers.

It was the second win this season against a Top 25 team -- the first time any Memphis team has ever done that -- and it gives the Tigers a real shot at making the American Athletic Conference championship game.

So, believe me, there is nothing I would like better than to write a column celebrating that victory and the players — especially on the previously-embattled defense — who made it happen.

But how do you fully celebrate when a player who was supposed to suit up for Memphis Saturday may have raped someone the night before?

The first obligation of a football program — and it’s unfortunate to even have to type this — is to recruit players who will not endanger members of the community.

Twice in the past three weeks, there is evidence that Memphis may have failed in that regard.

On Sept. 29, Memphis safety Shaun Rupert was charged with aggravated robbery, aggravated burglary and possession of a firearm during commission of a dangerous felony in April.

According to a police report, Rupert and another man pushed their way into the apartment of a another Memphis student, held the student at gunpoint and stole the student’s two watches, an Xbox and an Apple Macbook. The men then allegedly forced the student to get into the bathtub and told him, “If you call the police, there will be consequences.”

That was a black eye to the program. Rupert had been recruited to Memphis after being dismissed from Missouri in the wake of misdemeanor drug charges.

Memphis doubtless did its “due diligence” before accepting Rupert as a transfer.

Sometimes, that isn’t diligent enough.

Indeed, Suttles was also dismissed from his previous school. He was bounced by Nebraska in 2013 after getting into an altercation with a teammate.

Suttles then transferred to Memphis and emerged as a widely-admired team leader, a player that former coach Justin Fuente invariably included on the panels he assembled to talk to incoming recruits. I once wrote a glowing column about Suttles, and the person he had evidently become.

“Every day he is the same guy," Fuente told me. "Whether it's on the field or off, there's a purpose to what he is doing at all times.”

So it came as a shock Saturday when Memphis first announced that Suttles had been suspended and “removed from campus” as a result of an off-campus incident, and when the MPD then announced that Suttles had been charged with rape.

According to the police, officers responded to St. Francis Hospital at 3:15 a.m. Saturday for a reported assault. The incident occurred in the 3800 block of Douglas Avenue near the Memphis campus, on Thursday. The alleged victim told police she had been in a relationship with Suttles for approximately two months.

And, yes, it is important to note that both Suttles and Rupert are innocent until proven guilty, and that Suttles -- through his attorney, Blake Ballin -- specifically "denies the accusation of rape."

But it is just as important to recognize that two felony charges in three weeks is at the very least a troubling trend, and a reason to make certain that the football program isn't recruiting players who are putting other members of the community at risk.

By all accounts, coach Mike Norvell is a man who cares deeply that his players are good citizens. He runs a program that is known for its openness precisely because -- as he said during his impassioned remarks after the Navy game -- it has nothing to hide. Beyond all that, it was Fuente, not Norvell, who recruited Rupert and Suttles to Memphis in the first place. Those judgment calls were made before Norvell arrived.

But football can obscure anyone’s judgment. That was reaffirmed Saturday afternoon. As my colleague, Mark Giannotto, tweeted out updates to the Suttles story, a Memphis fan responded: “Enough, there is a game going on.”

And so there was. The home team won it. That’s to the credit of all involved. But on this day, given the news from the MPD, it was a good deal harder to celebrate.

Memphis player charged with rape, is kicked off team

MEMPHIS, Tenn. (AP) Police say Memphis defensive end Ernest Suttles has been charged with rape after an incident near campus, and Suttles was dismissed from the team before the Tigers played Navy on Saturday.

The Memphis Police Department wrote on Twitter that Suttles was charged after officers were called to a location near campus at 3:15 a.m. Saturday. Online records show he was booked into jail Saturday afternoon.

School officials released a statement Saturday morning saying the 23-year-old Suttles had been suspended for violating team rules for an off-campus incident. After police announced Suttles had been charged, school officials said the redshirt senior had been dismissed and removed from campus.

Suttles had been listed as a co-starter against Navy. He transferred to Memphis before the 2013 season after Nebraska kicked him off the team for allegedly hitting a teammate in the head with a bottle.

Nelly Rape Accuser Drops Charges

A woman who accused rapper Nelly of raping her on his tour bus near Seattle last week has decided to drop her claims. The woman’s lawyer, Karen Koehler, announced late Friday that her client wanted to stop the investigation and has refused to testify against Nelly in court. She “wishes she had not called 911 because she believes the system is going to fail her,” Koehler said. Nelly, whose real name is Cornell Iral Haynes Jr., was booked in Auburn, Washington last Saturday on suspicion of rape. He was later released and has not been charged with a crime.

SEATTLE — A woman who said the rapper Nelly raped her on his tour bus in a Seattle suburb last weekend is dropping her pursuit of criminal charges.

News outlets report that lawyer Karen Koehler said in a statement Friday that her client wanted to stop the investigation and would refuse to testify in court.

Koehler says the woman "wishes she had not called 911 because she believes the system is going to fail her."

Auburn police arrested Nelly, whose real name is Cornell Iral Haynes Jr., early Saturday in his tour bus at a Walmart. He was booked into jail on suspicion of second-degree rape and released later that day.

He has not been charged with a crime.

Nelly's attorney, Scott Rosenblum, has called the rape claim a "completely fabricated allegation."

More than 500,000 Rohingya flee from rape, fire and murder in Myanmar

HARI SREENIVASAN: During the past six weeks half a million Muslims from Myanmar known as the Rohingya have fled across their countries border into Bangladesh. The exodus comes after witnessing or experiencing horrific atrocities by government soldiers who rampaged in their villages as part of a crackdown on the long-persecuted ethnic minority. New York Times reporter Jeffrey Gettleman has reported vividly on the range of refugees, the widespread burning of homes the systematic raping of women and brutal killings of men and children. Gettleman is a Pulitzer Prize winner and currently the South Asia bureau chief. He joins me now via Skype from Dubai in the United Arab Emirates.

So first of all, for people who haven’t read your story, you end up helping us personalize this through one particular tale of a woman named Rajuma. Kind of summarize that for us.

JEFFREY GETTLEMAN: Yeah, this was this was one of the most upsetting stories I’ve ever done and I’ve been covering conflicts for 20 years and I was deeply disturbed and felt really helpless working on this story.

So I centered a piece on a young woman named Rajuma who had watched her village burn down in front of her eyes, was captured by government soldiers went back along with many other civilians and she told me that the soldiers separated the men from the women, they methodically executed all of the men and then began to rape the young women and Rajuma had a toddler boy in her arms and she said the soldiers ripped the little baby out of her arms, threw him into a fire, and killed him right in front of her and then proceeded to rape Rajuma and then left her in this burning house to die. And it was only a miracle when she woke up smelling smoke, ran out, naked, covered in blood, hid in a field and then kept running for the next three days until she got to the border of Bangladesh.

HARI SREENIVASAN: Well what’s remarkable about your story is, besides the fact that you verified all this from other witnesses and so forth, is that unfortunately her story is not an anomaly. You’re kind of trying to describe something that’s on a scale that we haven’t really heard about.

JEFFREY GETTLEMAN: Yes. This is just it. I mean I was I was in these camps in Bangladesh where half a million people have arrived nearly overnight fleeing these atrocities in Myanmar. And everywhere you go people have these stories to share, that are so disturbing. And the more you listen, the more you understand that the Rohingya have been demonized and dehumanized for decades. Like many people in Myanmar refer to them as insects or vermin. And then there was this official policy that they weren’t citizens, they didn’t really count. They were the most lowly people in the entire country. So then in August you had this attack by Rohingya militant groups against government positions and this became a long-awaited excuse for the military to just wipe out these people and they went village to village burning entire villages to the ground. More than 250 of them. And then they began to slaughter anybody they could get their hands on.

HARI SREENIVASAN: You know as these stories get documented as people start keeping track of, whether you want to call it a humanitarian crisis, crimes against humanity, a war crime, etc., what has the government response been?

JEFFREY GETTLEMAN: The Myanmar government has been denying that anything horrible happened and has called these clearance operations and said that the government troops were just trying to eliminate militants. But a half a million people fled. And they only did that because they felt like they would have been killed, had they stayed. And so many people told me these firsthand accounts, people from different villages, from hundreds of miles apart. And there was a distressing harmony to the accounts. And so you have this huge population that is now trapped in this border area not allowed to leave, not welcome by either side. When in these camps it was just this sea of plastic shelters as far as you could see to the horizon set up in these muddy hillsides, with just tons of people packed together, with very little food or shelter, and they all have been traumatized.

Actress Lysette Anthony claims Harvey Weinstein raped her at her home in the 80s. A new interview details the alleged attack, which the 54-year-old labelled ‘revolting’ and ‘pathetic. In the interview with The Times, she claims he showed up at her home in the late 1980s at 10am and pushed his way inside while she tried to fight him off. She told the London Times: ‘He pushed me inside, rammed me up against a coat rack. He was trying to kiss me and shove me inside.’ She says she pushed him away but he was too heavy and ‘finally I just gave up.’

Lysette said the attack was ‘revolting’ and left her ‘disgusted and embarrassed’. Scotland Yard were said to be investigating her claims last week. The board for the Academy of Motion Picture Arts and Sciences voted ‘well in excess of the required two-thirds majority’ in favour of expelling him, according to the BBC. Tom Hanks, Steven Spielberg and Whoopi Goldberg are some of the names on the 54-member board. In a statement, the Academy said: ‘We do so not simply to separate ourselves from someone who does not merit the respect of his colleagues but also to send a message that the era of willful ignorance and shameful complicity in sexually predatory behaviour and workplace harassment in our industry is over.’ It added: ‘What’s at issue here is a deeply troubling problem that has no place in our society. The Board continues to work to establish ethical standards of conduct that all Academy members will be expected to exemplify.’

One of Harvey Weinstein’s former employees has accused the movie mogul of raping her in the basement of his London offices, t he Daily Mail reported.

The ex-Miramax employee said she was so traumatized by the attack that it took her until last week to tell her husband about it.

“I just felt mortified and ashamed — and that no one would believe me,” she told The Mail on Sunday.

“He was incredibly well-connected, powerful and important — and I was just a nobody.”

Weinstein’s accuser has changed her name to “Sarah Smith” in order to protect her identity.

She said the assault took place in 1992 at the Miramax offices in Fulham in West London.

Smith, now in her 50s, said, “I was there on my own when he came to the office. He went down to the basement and he called my name. I went down — he was standing there with nothing on. I started to back away.”

She recalled being terrified by his strength and size.

“He grabbed me and he was so big and powerful. He just ripped my clothes away and pushed me, threw me down. Then...I kept shouting, ‘No! Stop!’ and tried to push him off. But he forced himself on me,” she said.

She recalls saying, “No!” over and over again.

“I was very aware that if a woman says no, it means no. And that was the one thing going through my mind throughout, ‘No, no, no, no!’” she said.

She remembers the encounter being over quickly, before he told her to “get out!”

“I remember walking home that night and it was cold and sodden. I was mortified and ashamed. I didn’t tell anyone,” she said.

“We live in more enlightened times now, but back then I just thought no one would believe me.”

She said she still suffers emotionally from the encounter.

“Even after all these years, I can still wake up screaming.”

Smith’s accusations come on the heels of an English actress’s allegations that Harvey Weinstein raped her in her London home in the 1980s.

Lysette Anthony, 54, reported the claim to Scotland Yard Wednesday.

“Have just reported an historic crime to @MerseyPolice...feel sick..so sad..” she wrote on Twitter.

Metropolitan police said Thursday that it was investigating Weinstein over claims that he had sexually abused an actress in London in the 1980s. They did not identify her.

Anthony described the encounter to the Times of London.

She said Weinstein showed up unannounced at her apartment one morning.

“He was trying to kiss me and shove inside me,” Anthony said. She said she tried to restrain him but he was too heavy.

“Finally I just gave up,” the actress reportedly said.

Anthony described the assault as “pathetic” and “revolting” in an interview Saturday.

She said she felt “disgusted and embarrassed.”

He wanted to leave it so badly but there were so many fears in his heart. He used think about necessities and luxuries. If he had to leave this hell called modern life he needed to be ready for a life which may not offer him any luxuries. And this was the fear holding him back. But this prison was suffocating him every day. He couldn't breathe any more. He wanted to be free.

What should he do? should he just leave and not think about any thing? Will it prove to be a huge mistake?

So one day he thought let's make a list of things he needs just for survival , and he took a pen and paper and started writing things down, after a long thinking he found out that all the necessities and luxuries of life are derived from following necessities :

The tanker had an accident in the morning when it overturned while taking a turn, because of the overturn the oil started spilling. seeing all this petrol the locals gathered around to collect what ever petrol they could find and eventually some thing sparked a fire and did so much damage.

It's sad to see how people gathered to steal that petrol. It's a problem with subcontinent people. We are always hungry. How much petrol they could have stolen from that tanker, at most 10-20 litres. But was that worth there life? The westerners might think that those people were so poor that's why they were trying to steal the petrol. Which isn't the case at all. In subcontinent we have this habit of being a show off or clever guy, those kind of clever heroes that we see in our movies, we try to be that guy. We love to break the lines. We are so happy when we borrow something from some one and then never return. We are that kind of society morally. No wonder we are where we are.

Most of the people think that the government is corrupt but its actually every one of us. Our labourer is corrupt, our shopkeeper is corrupt our street hawker is corrupt , and then we want to target Nawaz sharif only. Its a shame.

Who ever governs us is always going to be one amongst us. Aren't most of us corrupt. We are so happy when we steal some one else's money.

For Pakistan or India to prosper we need honest people in the lower classes and middle classes, not the other way around.

In this oil tanker incident , no one should have gone near the tanker . They were simply stealing. And in this race of dumbness they didn't even think that they were sitting on a mountain of fire which could go off any time.

A day after Pakistan said it wanted an exclusive dialogue with India on Kashmir, India responded saying it was ready for talks - but only on "relevant issues" that include cross-border terrorism and follow-up on the terror attacks in Mumbai and Pathankot.

"India would welcome a dialogue on contemporary and relevant issues in India-Pakistan relations," said External Affairs Ministry spokesperson Vikas Swarup.

The list, India said, will also include "infiltration of terrorists like Bahadur Ali, incitement to violence and terrorism across the border, parading of terrorists like Hafiz Saeed and Syed Salahuddin, and follow up on the Mumbai attack trial and the Pathankot attack investigation in Pakistan."

India has spoken repeatedly on the need to control cross-border terror from Pakistan.

Today, underscoring that addressing terrorism is "central to our engagement", External Affairs minister Sushma Swaraj said, "Unlike in the past, we can't agree that dialogue with sponsors and supporters of terrorism should carry on without being linked to action in that regard".

Pakistan's invitation for talks came as Prime Minister Narendra Modi, at an all-party meet on Kashmir yesterday, said, "Pakistan Occupied Kashmir is ours" and blamed the current unrest in Jammu and Kashmir on cross-border terrorism.

While Pakistan denied that terrorists were crossing over to India, investigators have said the revelations of arrested Pakistani terrorist Bahadur Ali point to the neighbouring country's involvement in the ongoing unrest in the state.

Bilateral dialogue with Pakistan was suspended after a group of armed terrorists attacked the Indian air base at Punjab's Pathankot in January. After its team visited the air base, Pakistan said India has failed to provide enough evidence that the attack was carried out by terrorists from across the border.

Last month, after Pakistan eulogized terrorist Burhan Wani, whose killing on July 8 had unleased the five-month long unrest, Ms Swaraj, in a sharp rebuke, said Pakistan's stated goal of detaching Kashmir from India will "not be realised to the end of eternity".
