<p>Since I didn&#8217;t get the ABC or NBC signal, I decided to improve the original one. <strong>Here is the updated and portable version.</strong><br />

<p>Please notice that I use a cardboard covered with aluminum foil as the reflector. I modified a paper clip (pointed by red arrow) to secure the antenna and the reflector.<br />

<p>Also, because the connection &#8220;wire&#8221; is much thicker, all the Vs are actually tilted forward, which seems like not affecting the signal strength.<br />

<p><strong>The direction is really important!</strong> It is one reason I modified the original one to this portable version. With one direction, I got several extra channels: 20-1, 20-4, 32-1, 32-2, 32-4, 32-5, 36-1, 36-2, 36-3. However, I lost 5-1, which is CBS and comes back when I turn the antenna set. I still didn&#8217;t have 11-1 (NBC) and 7-1 (ABC). <em>Will try during day time and see &#8230;</em></p>
<p>The other reason I modified my wire version is I was hoping to improve the signal by making the exact &#8220;clothes hangers&#8221; version. Because as shown in the commercial DB4 multi-directional HDTV antenna picture, the connection lines for the Vs are made of solid material not wire.<br />

<p>However, based on my test, changing the connection from thin wire into thick &#8220;clothes hangers&#8221; does NOT improve the signal. It suggests that only the Vs are the critical elements in receiving TV signal. <em>Good night!</em> </p>

<p>The oscar night motivated me to setup my HDTV. <em>I got my 1080p HDTV sitting there just as the monitor for Wii &#8211; what a waste @#$%</em> Since the cable service is expensive, I choose to use TV antenna to receive the free TV signals.</p>
<p>If it was ten years ago, I would go to a shop and buy one, maybe go to a library to check some reviews on different designs. <strong>But now, with google, I search!</strong> After some web reading, I decide to build one by myself. <em>The reasons are:</em></p>
<ul>
<li>It is very easy and much cheaper to build one</li>
<li>The quality of home-made version is better than some commercial ones</li>
<li>I want the TV working right now; cannot wait for the shipping &#8230;</li>
</ul>
<p>The first two hits on google about &#8220;<em>how to DIY antenna HDTV</em>&#8221; are <a href="http://www.tvantennaplans.com/">http://www.tvantennaplans.com/</a> and <a href="http://uhfhdtvantenna.blogspot.com/">http://uhfhdtvantenna.blogspot.com/</a>. They use the same design &#8211; modified DB4. <em>It is an very easy choice for me to follow them.</em></p>

<p>I made it in <strong>3 hours</strong>, including 30-min walk/searching in the hardware store to get the <em>BV-19 indoor VHF transformer</em>. My version is the method described in the youtube video with some modification. The cost is <strong>$1.14</strong> for the transformer, since I have all the other goodies. <em>Starting from the scratch, all the material costs less than $10.</em></p>

<p>It is very simple to make. Just follow the youtube video embedded above. Instead of using cloth hangers to link the Vs, I used wire and some soldering. I put a screw on the back of the wood to help mount it to the wall. I use aluminum foil covered wall as my reflectors. <em>Please notice that in the youtube video, the author didn&#8217;t put the reflectors. Using it significantly increases the signal.</em></p>

		<title>Image EGF receptor dimerization and activation in real-time by quantum dot and TIRF</title>

<strong>Preparation of Fab-QDs and tagging single EGFRs with the conjugate</strong></p>
<p><strong>AntiEGFR–Fab-biotin</strong> was produced by pepsin digestion of an anti-EGFR IgG (AB-11; clone 199.12, LabVision) followed by reduction of the resulting F(ab′)2 to yield a Fab′ fragment. The Fab′ was then conjugated with biotin through the unique -SH at its carboxy-terminal end. To prepare <strong>antiEGFR–Fab-QD conjugates</strong>, a solution of 0.7 nM antiEGFR–Fab-biotin in 1 ml of DMEM/F12 was added dropwise to 1 ml of 2 nM QD605-streptavidin conjugate (Invitrogen). Cell monolayers (50% confluent) previously serum starved for >1 h were incubated with 500 μl of the final antiEGFR–Fab-QD solution at 25 °C for ~2 min. The cells were then washed twice with PBS at 25 °C, placed in serum-free medium, and loaded onto the microscope stage at 37 °C in 5% CO2 for imaging.</p>
<p><strong>Imaging system and analysis</strong></p>
<p>Total internal reflection fluorescence microscope (TIRFM) imaging was performed using a Nikon Eclipse <strong>TE2000</strong> inverted microscope with <strong>100×/1.49NA Plan Apo objectives</strong> (Nikon). The sample was illuminated by the 488 nm line of a solid-state laser (Andor Technology) and images were acquired by the <strong>iXon back-illuminated EMCCD camera</strong> (Andor Technology). Individual trajectories of antiEGFR–Fab-QD were constructed by manual tracking plug-in in Image-J in parallel with the tracking module in Imaris (Bitplane). <em>10Hz, 100 ms per frame, not that fast.</em>

<p><strong>Couple interesting points in the paper:</strong></p>
<ul>
<li>&#8220;We tagged a small fraction of individual EGFR molecules with a conjugate of one CdSe quantum dot linked to one anti-EGFR antibody Fab fragment (antiEGFR–Fab) (Fig. 1a).&#8221;</li>
<li>&#8220;Single antiEGFR–Fab-quantum-dot (QD)-bound EGFRs (EGFR–Fab-QD) were visualized by total internal reflection fluorescence microscopy illuminated for <10 min to minimize the appearance of prolonged quantum dot dark states"</li>
<li>&#8220;Assuming monomer–dimer equilibrium, the Kd values for DelC and DelD were 2.9- and 14.2-fold greater than wild type (,10% errors; Supplementary Information 3). Thus, the structural stability of spontaneously formed dimers was also derived largely from the dimerization arms.&#8221;</li>
<li>&#8220;Thus, although ligand was necessary for receptor activation 5,6, it did not stabilize the dimer state27, contrary to the conventional view.&#8221;</li>
<li>&#8220;The frequency of spontaneous dimerization increased with receptor expression and was also location-dependent in the membrane of EGFR-overexpressing cells (A431 and BT20 cells; Fig. 3a).&#8221;</li>
<li>&#8220;The peripheral enrichment of both dimerization and ligand binding suggested that EGF bound preferentially to dimers 12,14.&#8221;</li>
<li>&#8220;For more than two decades, ligand binding and receptor dimerization were known to be critical steps in the EGFR activation process, but the exact relationship between these events was uncertain30. By analysing single receptors on living cells, we demonstrated that un-liganded EGFR fluctuates continuously between monomer and dimer states. Pre-formed dimers are primed for ligand binding and signaling, and in certain cell lines the distribution of dimers, and thus receptor responsiveness, are asymmetric. This asymmetry is related to both receptor expression level and the organization of the actin cytoskeleton 28,29.&#8221;</li>

<p>Epidermal growth factor receptor (EGFR) is a type I receptor tyrosine kinase, the deregulation of which has been implicated in a variety of human carcinomas. EGFR signalling is preceded by receptor dimerization, typically thought to result from a ligand-induced conformational change in the ectodomain that exposes a loop (dimerization arm) required for receptor association. Ligand binding may also trigger allosteric changes in the cytoplasmic domain of the receptor that is crucial for signalling. Despite these insights, ensemble-averaging approaches have not determined the precise mechanism of receptor activation in situ. Using quantum-dot-based optical tracking of single molecules combined with a novel time-dependent diffusivity analysis, here we present the dimerization dynamics of individual EGFRs on living cells. Before ligand addition, EGFRs spontaneously formed finite-lifetime dimers kinetically stabilized by their dimerization arms. The dimers were primed both for ligand binding and for signalling, such that after EGF addition they rapidly showed a very slow diffusivity state that correlated with activation. Although the kinetic stability of unliganded dimers was in principle sufficient for EGF-independent activation, ligand binding was still required for signalling. Interestingly, dimers were enriched in the cell periphery in an actin- and receptor-expression-dependent fashion, resulting in a peripheral enhancement of EGF-induced signalling that may enable polarized responses to growth factors.</p>

<p>I went to Heavenly last Saturday, with my Vado HD cam and, more importantly, a home-made attachment on my ski pack to mount the cam. With that design, I am able to video taping while skiing!!</p>
<p>Thanks for the <a href="http://www.elnino.noaa.gov/">El Niño</a>. This season has been <strong>EPIC</strong>. It was not snowing on Friday, but the temperature was low. The snow was stayed in powder status and waiting for me&#8230; It was cloudy and a little snowing after lunch. Anyway, the snow condition was great!</p>
<p>I went up and had my first couple runs below the <strong>Powderbowl express</strong>, along Powder Line and Fall Line. Then, I took the <strong>Canyon Express</strong> and had couple runs at High Roller. The <strong>Sky Express</strong> took me to the top of the mountain <em>(the lifting process was taped and in the first part of the video)</em>. I dropped down to the Nevada side through Skyline Trail, Milky Way, Dipper Woods and Galaxy. Following tips from <a href="http://www.tahoestash.com/">http://www.tahoestash.com/</a>, I had a great time in Galaxy Woods, where I had my footprints over the untouched powder and was able to capture the moment I fell into and buried in the fresh snow <em>(in the first part of the video, around 9&#8242;)</em>. After having several runs at Aries Woods and Little Dipper, I had my lunch in the East Peak Lodge. It started snowing in the afternoon, and I spent most of the time over the California side. Through Sky Express, I had great runs at Ellie&#8217;s and Pinnacles, which were newly groomed, and of course <strong>the woods between them</strong>!</p>
<p>The video was captured at 30 frames per second. Processing with Sony Vegas. I had to export the video as 60 frames per second to make it look &#8220;smooth&#8221;. Youtube only allows 10&#8242; per upload. I have divide it into to two: <strong>Part I</strong> includes Sky Express ride, Pinnacles run, Galaxy life and the fall; <strong>Part II</strong> starts from the fall and has my tree skiing in Galaxy Woods. <em>Enjoy!</em></p>

<p>Initially, I encoded the video as 30 frames per second, which is the one below. As you can see, it is pretty jumpy. <em>Do I need to buy a high-speed cam for my next skiing trip? $@$@$@$@</em></p>

<p>Of course, log out the current the user is the easiest way to delete all cached credential of shared network. However, when you don&#8217;t the condition to perform a logout, <strong>net use * \d</strong> becomes handy!</p>
<p>The <strong>IPC$ share</strong> is used with temporary connections between clients and servers by using named pipes for communication among network programs. It is primarily used for to remotely administer network servers. I tried to use <strong>net use z: \\ServerName\$IPC /delete</strong>, but got error message &#8211; the solution from google search is &#8220;I have to logoff&#8221;! Any suggestion? <em><a href="http://support.microsoft.com/kb/314984">Windows right-click trick</a> did not work &#8211; I cannot find the &#8220;Stop sharing&#8221; item &#8230;</em><br />
