BIOTECH INDUSTRIAL TRAINING PROGRAMME (BITP) 2013-14

On behalf of Department of Biotechnology (DBT), Ministry of Science and Technology, Government of India, Biotech Consortium India Limited (BCIL) is implementing since last 18 years, a scheme of practical industrial training for M.Sc./M.Tech./B.E./B.Tech. Biotechnology students. The objective of this programme is to provide industry-specific training to Biotech students for skill development and enhancing their job opportunities in biotech industry. This programme provides an opportunity to Biotech Industry for training and selecting suitable manpower. The training period is for six months during which a trainee is paid stipend of Rs.8000/- per month and trainer company is paid a bench fee to cover the expenses for providing training. The next batch of training will commence in October.

Applications are invited for the Award of UGC BSR Research Fellowship in Sciences for Meritorious Students (Botany). Under this scheme the UGC has approved 05 (Five) Research Fellows at a monthly Fellowship along with a contingent grant as per detail given below.

1. For Non UGC/CSIR/SLET/GATE/ (JRF) Examination and P.U. Ph.D. Entrance Test Qualified candidates, the Fellowship will be Rs. 14,000/- p.m. (fixed) plus H.R.A. for 1st and 2nd year and Contingency @ Rs. 12,000/- p.a. as per UGC/University rules. In the 3rd,4th and 5th year fellowship @ Rs. 16,000/- p.m. (fixed) plus H.R.A. and Contingency @ Rs. 25,000/- p.a. as per UGC/ University rules.

Qualifications/ Eligibility:

The candidates should be M.Sc. in Botany with at least 55% marks. The candidates who are enrolled/registered or eligible for enrolment/registration for Ph.D. in the subject of Botany as per the P.U. guidelines and have passed entrance test of Panjab University or UGC/CSIR/SLET/GATE/ (JRF) Examination are eligible.

Preference for selection will be given to those who are not getting fellowship but are otherwise eligible for the grant.

Selected candidates who are eligible for enrolment will have to submit duly completed enrolment form within three days of the selection failing which the selection shall get cancelled without any prior notice.

The selection will be subject to the approval by the UGC. The Department of Botany or the University will not be responsible for the award of fellowship in case UGC, for whatsoever reason/s, fails to approve or delays in approving or granting the fellowship. The department/ university is only the recommending body.

The allotment of guide and field will be subject to availability of seats with the guide/in the lab.

Tenure: The tenure of fellowship is initially for two years under the RFSMS scheme. Upon expiry of this period, the work of the Fellow will be evaluated by Experts Committee of the university. If the research work is found satisfactory, his/her tenure will be extended for a further period of three years. The total period of fellowship is 5 years with no further provision of extension.

The selected candidates shall be entitled to the fellowship only after approval by the UGC and receipt of the grant.

About MGDC

MGDC is at mission to motivate students, teachers and scientists in India to take to “alternatives” in the place of animals in life science / biomedical science education, research, and risk assessment, along the lines of 3Rs principle, and to impart training in the “alternatives”.

Background of the workshop

Advancements in cell culture and in vitro toxicology techniques have given researchers a viable alternative and/or complement to live animal testing. These methods can be used to elucidate the mechanism of toxic action of chemicals on particular target organ/cells, and various toxicity endpoints can be determined at levels including cellular, molecular and organ level. A variety of cell-based methods including monolayer cultures (2 Dimensional), multilayered cultures (3D), miniaturized 3D models and co-culture models have been developed and used for the assessment and/or ranking of various chemicals/drugs.

The recent tissue engineered/reconstituted 3D in vitro models, in which stem-cell derived cells are grown within or on extracellular matrix gels where cells behave as parts of whole living organs, overcome the limitations in the monolayer/2D cultures. Several such 3D models that have been brought into use include 3D hepatocyte cultures, tumor models, cardiac tissue models, bone tissue models, cornea tissue models, and skin models, etc., the use of which can save millions of animals from pain and distress. More importantly, the data generated are human-relevant. Particularly, 3D epithelial models can completely replace animals in cosmetics testing. The SkinEthic from France, a pioneer in developing and distributing in vitro human skin & epithelial models, joins hands with MGDC to educate / train the Indian academic, scientific and regulatory communities in alternatives to animal testing using SkinEthic’s in vitro reconstructed 3D models in a hands-on training workshop at MGDC, Tiruchirappalli. Nominations/Registrations are invited.

The workshop will cover

· Presentations on alternative methods, 3D tissue engineering, the various reconstituted 3D tissue models, etc.

· Lab exercises on in vitro skin irritation protocol

· Trainees will perform hands-on application of test substances on real tissue-engineered models, generate data and interpret the results

· This training session is a unique opportunity to acquire the most advanced techniques – including tips and tricks – compliant with the 7th Amendment of the EU Cosmetics Directive, the REACh program, part of OECD guidelines, and DCGI’s directive IS4011

Who can participate?

· Faculties and Scientists from Universities and Research Institutes, with a background knowledge of animal cell / tissue culture

· Representatives from Regulatory Bodies /Authorities / Science Academies

· Enrollment will be limited to 8 participants

Registration Fee: Rs. 25,000 /-

This highly subsidized registration fee includes guest house accommodation where rooms need to be shared, and reasonably good food for the 2 days. Research into Cancer–Immune System Interactions

Immune cells can infiltrate the cancer microenvironment, the consequences of which are not well understood. The multi-faceted immune presence interacts with cancer cells in inhibitory and/or stimulatory ways, resulting in complex cancer–immune interactions. Below we describe our current mathematical and computational approaches to understand the consequences and implications of these intercellular interactions.

Tumor-Promoting Inflammation

The presence of cancer within a host initiates a systemic immune response towards the transformed cells. Inflammatory immune cells such as neutrophils, platelets, macrophages, and natural killer cells, are recruited to the tumor site where they initiate the wound healing process. Tumors, sometimes viewed as wounds that never heal, can be promoted by these inflammatory actions. Once the adaptive immune response is activated by dendritic cells and macrophages, CD8+ T cells, or

cytotoxic T lymphocytes, infiltrate the tumor and induce apoptosis in the target tumor cells. Depending on the cytokines and other signals present in the tumor microenvironment, recruited immune cells will either form a pro-tumor immunity (typified by cytokines such as TGF-β, IL-6, and IL-10 and cells such as M2 macrophages, Th-2 T helper cells, and myeloid derived suppressor cells) or an anti-tumor immunity (typified by cytokines such as IFN-γ, IL-2, and IL-12 and cells such as M1 macrophages, Th-1 T helper cells, and cytotoxic lymphocytes).

To investigate the role of tumor-promoting inflammation, an emerging hallmark of cancer, we have developed a mathematical model for cancer-immune interactions that can capture both the pro-angiogenic, tumor-progressing actions of a pro-tumor inflammatory microenvironment, and the anti-angiogenic, tumor-inhibiting actions of an anti-tumor inflammatory microenvironment. This model utilizes principles of generalized logistic growth, which captures some of the inherent variability underlying tumor growth in an immune competent host that is often neglected in macroscopic measurements and in mathematical models. From model simulations, the two types of inflammation (pro-tumor or anti-tumor) resolve into two fundamentally different classes of outcomes, where inflammation-enhanced tumor progression must either result in a decreased tumor burden, as in the anti-tumor case, or in an increased tumor burden, as in the pro-tumor case. These results suggest that, in some cases, fast tumor growth may be advantageous, if it leads to a significantly smaller tumor burden. In such cases, it is possible that treatments should be targeted towards enhancing the stability of an anti-tumor inflammatory environment instead of towards immediate tumor regression.

Tumor Immunoevasion

Despite highly evolved adaptive immune responses, tumors often manage to escape recognition by the immune system. This process is known as immunoevasion, and is another emerging hallmark of cancer.

When hematopoietic stem cells leave the bone marrow they differentiate into either lymphoid progenitors or myeloid progenitors. Lymphoid progenitors migrate to the thymus where they differentiate into T, B, and NKT cells. Myeloid progenitors differentiate into monocytes, migrate to tissues, and differentiate into myeloid cells such as dendritic cells (DCs) and macrophages. When an immature DC encounters an antigen, it internalizes the antigen to display fragments on its membrane. The DC then matures as it migrates to a lymph node. Maturation involves the loss of ability to engulf pathogens and an increased ability to communicate with T cells. Within the lymph nodes (collection points where antigen presenting cells interact with T cells attracted to the node via chemotaxis), mature DCs activate naïve T cells to develop a specific immune response. Activated cytotoxic T cells undergo rapid clonal expansion and migrate throughout the body in search of relevant targets. T cells perform their cytotoxic function by inducing apoptosis in the target cell through the secretion of perforin and granzymes or through Fas/Fas-ligand binding.

Within the process of activating the adaptive immune response described above, two significant functions may be subverted by tumors: antigen presentation (maturation of DCs) and T cell functionality.

Antigen presentation suppression

If antigen presentation is blocked then naïve T cells are not activated and a specific immune response is not mounted. A number of cytokines, chemokines and growth factors, such as HIF-1α, VEGF, nitric oxide, and reactive oxygen species (ROS), produced within the tumor microenvironment may interfere with the process of DC maturation. Without DC maturation, there may be an accumulation of immunosuppressive factors, such as myeloid derived suppressor cells (MDSCs), in the tumor microenvironment resulting in immunoevasion.

In order to investigate this process from a modeling perspective, we formulated a system of ordinary differential equations in a predator-prey type system, where the prey (cancer cells) have a defense mechanism (immunoevasion) against recognition by the predator (immune system). Our analysis suggests that this mechanism can have significant effects on overall tumor-immune dynamics, ultimately allowing for either tumor suppression or tumor escape in a manner that depends on the strength of the immune suppression [Kareva et al, 2010]. Currently, we are investigating the possible role of glycolysis and the resulting reduction of pH in a hypoxic tumor microenvironment as another possible mechanism for immune evasion.

Impaired T cell functionality and immune resistance

The immune response poses a second barrier to tumor growth after the angiogenic switch. Immune surveillance of tissues allows for early detection of transformed cells. If these transformed cells are not recognized as “self”, they are eliminated by the immune system. Through repeated exposure of the transformed cells to this immune selection process, various phenotypes can arise within the cancer cell population, creating a heterogeneous population of neoplastic cells. These immunoedited cells may develop the ability to evade the immune response and grow in an uncontrolled manner.

After prolonged periods of immune-induced dormancy, T cells can lose effectiveness in their cytotoxicity. This loss may be due to either T cell tolerization to the cancer cells or to an increased cancer cell resistance to immune attack. Both of these mechanisms are intertwined in the process of immunoediting that can lead to tumor escape from immune control.

To investigate the heterogeneous population-level dynamics involved in this immune selection process, we are working on a mathematical model that can capture the essential cancer-immune interactions that may lead to T cell tolerization and / or the accumulation of immune-resistance by cancer cells. These two fundamentally different mechanisms of immune evasion would require specifically targeted therapies, which could be analyzed theoretically with this mathematical model.

Cancer Stem Cells and Immune System-Modulated Tumor Progression

The role of the immune system in tumor progression has been subject to discussion for many decades. Numerous studies suggest that a low immune response might be beneficial, if not necessary, for tumor growth, and only a strong immune response can counter tumor growth and thus inhibit progression.

Without an immune response, a heterogeneous tumor population comprised of cancer stem cells and non-stem progenitors grows as conglomerates of self-metastases [Enderling etal., 2009]. This morphological phenomenon results from the interplay of cell proliferation, cell migration and cell death. With increasing cell death intra-tumoral spatial inhibitions are loosened, which in turn enable cancer stem cell cycling and thus, counter-intuitively, tumor progression [Enderlinget al., 2009b]. By overlaying on this model the diffusion of immune reactants into the tumor from a peripheral source to target cells, we simulate the process of immune-system-induced cell kill on tumor progression. A low cytotoxic immune reaction continuously kills cancer cells and, although at a low rate, thereby causes the liberation of space-constrained cancer stem cells to drive self-metastatic progression and continued tumor growth. With increasing immune system strength, however, tumor growth peaks, and then eventually falls below the intrinsic tumor sizes observed without an immune response. Focusing only on the cytotoxic function of the immune system, we were able to observe all immunoediting roles of the immune system: immune promotion at weak immune responses, immunoinhibition at strong immune responses, and immunoselection at all levels. Simulations of our model support a hypothesis previously put forward by Prehn [Prehn, 1972] that comparable tumor sizes can be observed for weak and strong immune reactions. With this increasing immune response the number and proportion of cancer stem cells monotonically increases, implicating an additional unexpected consequence, that of cancer stem cell selection, to the immune response.

Cancer stem cells and immune cytotoxicity alone are sufficient to explain the three-step “immunoediting” concept — the modulation of tumor growth through inhibition, selection, and promotion. We propose more generally that a stem-cell-expansive influence may take the form of anything that encourages morphological fingering. Beyond immune response, this could include cell death, or even growth within restricted thin channels, as might be expected e.g. during invasion of host tissue.see more......

Human Genome Project is largely complete, so the problem facing by scientists is how to maximize the use of the newly acquired data for improving health care. It is estimated that the number of therapeutic targets available for drug discovery will increase from the current number of 600 to 1000 to perhaps as many as 5000 to 10,000. In addition to the challenges that this number provides to the pharmaceutical industry, there is the issue of sequence variation through Single Nucleotide Polymorphisms (SNPs), some of which may impact upon the way that the body handles drug treatment. This may be due to a direct effect on the binding site of the protein target through non-conservative alterations of the amino acid sequence, or else through indirect effects on drug metabolizing enzymes. In order to meet these challenges, the combination of structural proteomics and computer-aided small molecule design provides opportunities for creating new molecules in silico; these may be designed to bind to selected pharmacogenetic variants of a protein in order to overcome the non-responsiveness of certain patient groups to a particular medicine.

In silico drug discovery methods reduce both time and cost of development of novel drug/lead molecules. Knowledge about the molecule, its interaction with the drug, molecular modeling, and new drug development creates an awareness of the molecule at the organ level and aids in the prevention, diagnosis, and treatment of diseases. Computational tools have the advantage of delivering new lead candidates more quickly and at lower cost. In the 21st century, in silico methods facilitate target identification, structure prediction and lead/drug discovery. The computational methods expect more reliable and expeditious protocols for development of novel ideas, which increase the potential leads. The computer-assisted molecular design has succeeded in the QSAR and protein modeling algorithms to improve activity of lead compounds. Selecting candidates via the described in silico virtual screening, should help to reduce the list of candidate molecules and thereby reduce time and costs significantly.

The major challenge for science education today is the rapidly changing technology that needs to be integrated in the teaching–learning processes. Transfer of knowledge from research laboratories to educationists directly will be a correct step in this direction. The Science Academies’ Refresher Course on ‘Advances in Plant and Animal Biotechnology’ promises knowledge updates in plant tissue culture techniques, developing transgenics, molecular confirmation and transgene expression, primary stem cell cultures, bio-informatics and allied techniques. The two-week Course will be held from 3 to 16 February 2013 spread over a few institutions in Mumbai (IWSA, Vashi; ACTREC, Kharghar; Karmaveer Bhaurao Patil College, Vashi; Dr D. Y. Patil University, Belapur). It aims at providing hands-on-training in plant and animal cell handling, isolation of DNA and selection of vector systems for cloning, in addition to the harnessing of bioinformatics, knowledge of IPR, writing of project proposals and ethics in biotechnology. The course will also comprise of lecture-demonstration sessions on new generation sequencers, live animal imaging, confocal microscopy, etc.

Post-graduate teachers from Life Sciences who wish to participate in the Refresher Course may apply through proper channel with the following details – name, date of birth, gender, e-mail, official and residential addresses, telephone numbers, academic qualifications, courses taught, positions held and tenure. Submission of a brief statement (not more than 500 words) why they think the Course will improve their classroom teaching should also be sent. Applications should reach the Course Coordinator before 15 December 2012.

Selected candidates will be informed by the first week of January 2013 and outstation candidates will be provided accommodation, local travel and round trip

The National Center for Tumor Diseases (NCT) Heidelberg offers a highly interactive and collaborative scientific environment with a strong focus on translational cancer research. The laboratory of Prof. Stefan Fröhling in the Department of Translational Oncology is interested in the molecular pathogenesis of epithelial and hematopoietic malignancies, with the ultimate goal to identify novel therapeutic targets. Specific projects include the identification of essential proteins and signaling pathways in Cancers dependent on mutant KRAS, the most commonly mutated human oncogene, and the identification of genetic vulnerabilites in epithelial Cancers and acute myeloid leukemia using functional genomics screens. Applicants should submit the following materials: CV, letter of motivation including expected availability date, complete list of publications, certificates, 2-3 references.

- University degree in biology, biochemistry, genetics, or a related field

- High motivation and strong interest in Cancer biology

- Strong laboratory and analytical skills

Rajiv Gandhi National Fellowship for SC/ST Candidate For The Year 2012-13

The Rajiv Gandhi National Fellowship (RGNF) Scheme for Scheduled Caste and Scheduled Tribes is formulated and funded by Ministry of Social Justice & Empowerment and Ministry of Tribal Affairs. The scheme is open to candidates who belong to Scheduled Caste & Scheduled Tribe and wish to pursue higher studies such as regular and full time M.Phil. and Ph.D degrees in Sciences, Humanities, Social Sciences and Engineering & Technology. There are 2000 slots for Scheduled Caste and 667 slots for Scheduled Tribe candidates every year for all the subjects.

OBJECTIVE

The scheme has been initiated keeping in view the social background of the candidates for the deprived section of the society and to provide them opportunity to undertake advanced studies and research. The objective of this award is to provide fellowships in the form of financial assistance to students belonging to SC/ST to pursue higher studies leading to M.Phil and Ph.D degrees in Sciences, Humanities and Social Sciences and Engineering & Technology.

TARGET GROUP

SC/ST Candidates who have passed the Post Graduate examination in concerned subject and who desire to pursue research without JRF of UGC-NET or UGC-CSIR NET.

ELIGIBILITY:

The candidate should belong to SC/ST and should have passed the Post Graduate examination. The selection will be made on the basis of merit of the candidates. The candidate must get himself/herself registered for M.Phil./Ph.D at the first available opportunity but not later than within a period of two years from the receipt of award letter. This period is not extendable. However, the actual payment of fellowship will be made with effect from the date of approval of fellowship or date of joining whichever is later.

NATURE OF ASSISTANCE AVAILABLE UNDER THE SCHEME: The tenure of fellowship is initially for two years under the RGNF Scheme. Before expiry of this period, the work of the Fellow will be evaluated by a Committee of three members comprising of Head of the Department, Supervisor and one outside subject expert to be constituted by the concerned Department of the /University/Institution/College. If the research work is found satisfactory, his/her tenure will be extended further for a period of three years under the enhanced emoluments of the Rajiv Gandhi National Senior Research Fellowship (RGNSRF). The recommendation of the Committee shall be submitted to the UGC for upgradation to the level of RGNSRF. The work done and the time spent on scholarships or fellowships of any agency other than the UGC will not be taken into account while considering the proposals for enhancement in the value of fellowship. The fellowship may be withdrawn if the work is not considered satisfactory or the candidate fails in any of the examinations related to Ph.D. In case the work for the first two years is not found satisfactory, an additional year will be given to him/her for improvement. However, during this period he/she will be designated as Rajiv Gandhi National Junior Research Fellow. In such cases work will be evaluated again before third year of the tenure, and if improvement is found, the Fellow will get two more years under the RGNSRF. Thus, the total period of fellowship (RGNJRF and RGNSRF) is for five years, with no further provision of extension. The candidate would be eligible for following financial assistance:
